Effect of ibuprofen on hippocampal gene expression in APP-PS1 mice

Project description:NSAIDs (non-steroidal anti-inflammatory drugs) inhibit cyclooxygenase (COX) enzymes and prevent Alzheimer's disease (AD) at preclinical stages in cognitively normal aging populations. We modeled NSAID prevention of memory impairment in AD model mice to identify novel targets of NSAID action. We found that the widely-used NSAID ibuprofen prevented early hippocampus-dependent memory deficits in APP-PS1 mice. We therefore analyzed gene expression in the hippocampus of these mice. We treated male APPSwe-PS1deltaE9 mice (strain originally provided by Dr. David Borchelt and fully back-crossed to a C57BL/6J background) and their wild-type littermates with ibuprofen in chow from 3 to 6 months of age, after which we sacrificed the mice and dissected one hippocampus from each mouse for RNA isolation and microarray analysis. In each genotype-treatment group, we analyzed samples from 5 mice. Note: In our initial analysis, we found that sample GSM1644138, Hippocampus_WT_Con_rep5, was an outlier in overall gene expression. We therefore removed this sample before performing the analysis published in PMID 27190010.

Time-dependent olfactory proteomics in Tg2576 mice

Project description:Olfaction is often deregulated in Alzheimer´s disease (AD) patients, being also impaired in transgenic Tg2576 AD mouse model, which overexpress the Swedish mutated form of human amyloid precursor protein (APP). However, little is known about the molecular mechanisms that accompany the neurodegeneration of olfactory structures in Tg2576 mice. For that, we have applied proteome-wide approaches to probe molecular disturbances in the olfactory bulb (OB) dissected from Tg2576 mice respect to age matched wild-type littermates (2 and 6 months of age).

Differentially expressed miRNAs between Alzheimer's disease and control mice's hippocampus

Project description:MicroRNAs influence restorative physiological or degenerative pathological processes in several neurological disorders, yet their role in Alzheimer’s disease remains unclear. Here we compared microRNAs expression in the period of AD pathogenesis when mice first begin to have detectable extracellular Aβ deposition. Tg2576 mice aged 9-10 months and their wild type littermates, as well as a double transgenic model of AD, APP/ PS1M146V/+ and wild type C57BL/6J mice both at 3-4 age months (n=3) were used. Hippocampus tissue miRNAs were detected by hybridization to microarrays having 924 mammalian miRNA probes.The result indicated that some of the deregulated miRNAs in AD mice have been implicated in amyloid beta production.

Transcriptomic landscape of the hippocampus in 2- and 4-month-old rTg4510 mice and wild-type littermates

Project description:The objective of this study was to compare the hippocampal transcriptomic landscape of the rTg4510 mouse model of tauopathy (Tg) at two ages (2 and 4 months) with that of wild-type (WT) littermates at the same ages, and to assess age-related changes (2 vs 4 months) within each genotype. The rTg4510 line overexpresses a repressible form of human tau carrying the P301L mutation. Tau expression is controlled by a tetracycline-responsive element (tetO) and driven by a forebrain-specific transactivator under the CaMKIIα promoter. Mice were maintained without doxycycline. In the absence of doxycycline, tau expression is induced primarily in the hippocampus and cortex, leading to progressive tau pathology (including tau hyperphosphorylation and neurofibrillary tangle formation) and associated neurodegenerative phenotypes. Transcriptomic changes in the hippocampus during early disease evolution (2 vs 4 months) are still incompletely characterized. This dataset provides bulk hippocampal RNA-seq data to support the analysis of age- and genotype-associated molecular changes in this model.

Effect of chronic valproic acid treatment on hepatic gene expression profile in Wfs1 knockout mouse.

Project description:Aim of the present study was to compare the effect of chronic VPA treatment in wild type and Wfs1 knockout mice on hepatic gene expression profile. Wild type, Wfs1 heterozygous and homozygous mice were treated with VPA for three months (300 mg/kg i.p. daily) and gene expression profiles in liver were evaluated using Affymetrix Mouse GeneChip 1.0 st arrays. male wild-type and Wfs1 mutant (+/- and -/-) mice were treated with valproic acid (300 mg/kg/day i.p.) or vehicle (saline, 10ml/kg i.p.) for 12 weeks starting from age 4 to 6 weeks. At age 16 to 18 weeks mice were killed, RNA extracted from the liver and analysed using Affymetrix Mouse Gene 1.0 ST Arrays. Expression of few genes was verified using RT-PCR. There were 8 animals in every group, 48 animals total.

Sex differences in the molecular signature of the developing mouse hippocampus

Project description:We used LC-MS/MS based proteomics to identify changes in protein expression in the the female and male mouse hippocampus at 1, 2, and 4 months of age. This proteomics dataset was correlated with an accompanying mRNA dataset to identify genes that show mRNA changes in a different direction that their respective proteins. A modest correlation was found between mRNA and protein abundance in the mouse hippocampus (spearman rank coefficient = 0.53).

Transcription profiling of mouse APP transgenic mouse model of Alzheimers disease treated with BDNF

Project description:We examined transgenic (TG) mice expressing human APP695 bearing the double Swedish (671KM>NL) and Indiana (717V>F) amyloid precursor protein (APP) mutations. Lentiviral vectors constitutively expressing BDNF-GFP under control of the CMV/Ã-actin hybrid promoter or GFP alone were injected into the entorhinal cortices of TG mice bilaterally at age 6 months, a time point by which neuropathological degeneration and cell loss are established. Age-matched wild-type littermates underwent sham surgery or injection of lentivirus expressing GFP into the entorhinal cortices bilaterally. Experiment Overall Design: 26 Samples total: 4 biological replicates of APP transgenic mice BDNF treated, 4 biological replicates of APP transgenic mice GFP treated, 3 biological replicates of non-trangenic mice sham lesion and 2 biological replicates of non-transgenic mice GFP treated for both tissues: Entorhinal cortex and hippocampus.
