Project description:Drought priming is a promising approach to improve drought tolerance, and root apex plays important role in coping with drought adversities in plants. It has been observed that less inhibition of root growth under drought stress was found in paralleling with the enhanced drought tolerance induced by drought priming in wheat. However, the contribution and mechanisms of root apex to the improved tolerance induced by drought priming remains unknown. Here, the transcriptome and proteome of three different zones along the root axis under drought stress were investigated. The two zones distal from the root apex showed more sensitive to drought priming and the later drought stress than the zone proximal to the root apex, as exemplified by the principal component analysis of the different expressed genes (DEGs) among treatments. The DEGs which decreased by drought stress, while higher expressed in primed plants than non-primed plants, might play critical roles in enhanced root growth in primed plants. These genes were mainly involved in the pathways related to cell membrane sensing, plant hormone signaling, stress defense and cell more modification. Moreover, the plant hormone pathway showed positive correlated between transcriptome and proteome analysis, and genes network analysis hinted that ABA receptor and aquaporin could be potential markers in regulation of priming induced root growth and drought stress tolerance.

Project description:Drought is one of the most detrimental environmental factors that adversely affect crop production, thus jeopardizing food supplies for a growing world population. Over the past years, it has become evident that microorganisms associated with plants can enhance drought tolerance. However, the specific genetic and molecular mechanisms underpinning bacterial induction of drought tolerance in plants are still largely unknown.In our work, we have shown that a root endophytic Flavobacterium confers significant drought tolerance to Arabidopsis thaliana. We showed that the bacterium enhances drought tolerance by maintaining (and inducing) root hair formation and elongation under drought. By combining molecular, genetics, and omics technologies we identified an essential role of ethylene and two ethylene-induced transcription factors belonging to the ERF family (ERF115 and ERF114) through which flavobacterium exerts its effect on plant root hair formation and drought tolerance. Here we performed transcriptome analysis using RNA-seq to identify genes downstream of the two ERF transcription factors.

It was reported that DREB1A improves drought stress tolerance in various crops but causes dwarfism, and GA5 encodes a GA synthetic enzyme which enhances plant growth. We performed microarray experiments using a single overexpressor of GA5 and the double overexpressor of DREB1A and GA5 to study the potential interactions of the gene networks regulated by the two factors in the double overexpressor.

Genome-wide analyses revealed that OsNAC6 directly up-regulated 13 genes. Taken together, OsNAC6 is a valuable candidate for genetic engineering of drought-tolerant high-yielding crops.

Project description:Lamin proteins in animals are implicated in many nuclear functions, including chromatin organization, signaling transduction, gene regulation, and cell differentiation. Nuclear Matrix Constituent Proteins (NMCPs) are lamin analogues in plants, but their regulatory functions remain largely unknown. In this work, we report that OsNMCP1 positively regulates root growth and drought resistance through modulating chromatin state and expression levels of many genes functioning in root growth and stress resistance. OsNMCP1 is localized at the nuclear periphery and induced by drought stress. Overexpression of OsNMCP1 resulted in a deeper and thicker root system and enhanced drought resistance compared to the wild-type control. Assay for transposase accessible chromatin with sequencing (ATAC-seq) analysis revealed that OsNMCP1-overexpression altered chromatin accessibility in many genes related to drought resistance and root growth, including OsNAC10, OsERF48, OsSGL, and OsMSR15. Interestingly, OsNMCP1 can interact with SWITCH/SUCROSE NONFERMENTING (SWI/SNF) chromatin remodeling complex OsSWI3C. The reported drought resistance or root growth related genes that were positively regulated by OsNMCP1 were negatively regulated by OsSWI3C, and OsSWI3C overexpression led to decreased drought resistance. We propose that the interaction between OsNMCP1 and OsSWI3C may lead to release of OsSWI3C from the gene silencing complexes SWI/SNF under drought condition thus changing the chromatin accessibility in genes related to root growth and drought resistance.

Project description:Plant responses to drought stress require the regulation of transcriptional networks via drought responsive transcription factors, which mediate a range of morphological and physiological changes. AP2/ERF transcription factors are known to act as key regulators of drought resistance transcriptional networks; however, little is known about the associated molecular mechanisms that give rise to specific morphological and physiological adaptations. In this study, we functionally characterized the rice (Oryza sativa) drought responsive AP2/ERF transcription factor, OsERF71, which is predominantly expressed in the root meristem, pericycle, and endodermis. Overexpression of OsERF71 either throughout the entire plant or specifically in roots, resulted in a drought resistance phenotype at the vegetative growth stage, indicating that overexpression in roots was sufficient to confer drought resistance. The root specific overexpression was more effective in conferring drought resistance at the reproductive stage, such that grain yield was increased by 23-42% over wild type plants or whole-body overexpressing transgenic lines under drought conditions. OsERF71 overexpression in roots elevated the expression levels of genes related to cell wall loosening and lignin biosynthetic genes, which correlated with changes in root structure, the formation of enlarged aerenchyma and high lignification levels. Furthermore, OsERF71 was found to directly bind to the promoter of OsCCR1, a key gene in lignin biosynthesis. These results indicate that the OsERF71-mediated drought resistance pathway recruits factors involved in cell wall modification to enable root morphological adaptations, thereby providing a mechanism for enhancing drought resistance.
