TIMP-GLIA (gliadin nanoparticle) treatment in transgenic mice expressing celiac disease associated HLA-DQ8

Project description:This submission contains raw data obtained from RNA sequencing of mouse spleen cells. Spleen cells were derived from adult female HLA-DQ8, huCD4 transgenic Ab0 NOD mice, treated intravenously either with tolerogenic, immune-modifying nanoparticles containing gliadin (TIMP-GLIA), particles containing irrelevant control ovalbumin (TIMP-OVA), or not receiving treatment. Mice were then immunized with gliadin (except for negative controls), and spleens were collected after 28-30 days. Spleen cells were stimulated with gliadin (or controls) in culture for 72h, and RNA was obtained from cells for transcriptomic studies.

[E-MEXP-3786] IGF-I-induced chronic gliosis and retinal stress lead to neurodegeneration in an animal model of retinopathy

Project description:Transcription profiling by array of mouse male retinas to investigate IGF-I-induced chronic gliosis and retinal stress IGF-I exert multiple effects in different retinal cell populations in both physiological and pathological conditions. Transgenic mice overexpressing IGF-I in the retina showed impaired electroretinographic responses at 6-7 months of age that worsen with age. This retinal neuronal dysfunction was correlated with the loss of rod photoreceptors, bipolar, ganglion and amacrines cells. Neuronal alterations were preceded by the overexpression of retinal stress markers, acute phase proteins and gliosis-related genes. IGF-I overexpression leads to chronic gliosis and microgliosis in TgIGF-I retinas, with mild oxidative stress, impaired recycling of glutamate and defective potassium buffering. These impaired supportive functions can contribute to neurodegeneration in TgIGF-I retinas, together with the increased production of pro-inflammatory cytokines, potential mediators of neuronal death.

Project description:C57BL/6 mice were subjected to sham surgery or orthotopic Hepa1-6 hepatic tumor inoculation. Spleen tissues from mice subjected to sham surgery (control mice), mice bearing a 2-week orthotopic hepatoma, and mice bearing a 4-week othotopic hepatoma were collected. Total RNA was extracted from the spleen tissues, and we used the RT² Profiler PCR array-mouse common cytokines (SABioscience) array panels to quantitate gene expression of cytokine genes from the spleens.
