Project description:Multiple myeloma (MM), also known as plasma cell myeloma, is a cancer of plasma cells, a type of white blood cell normally responsible for producing antibodies. There is no cure for MM. In addition, the mechanism underlying abnormal production of plasma cells is not clear. In this experiment, peripheral blood was obtained from normal healthy donors and multiple myeloma (MM) patients. Peripheral blood mononuclear cells (PBMC) were separated by Ficoll separation solution. Samples of four donors were pooled and Samples of four MM patients were pooled. The aim was to characterize the mRNA profile of MM patients compared to healthy donors and find the new target of diagnosis or treatment for MM.

Project description:MicroRNAs are small regulatory RNAs that are deregulated in a wide variety of human cancers, including different types of B-cell lymphoma. Nevertheless, the feasibility of circulating microRNA for early diagnosis of B-cell lymphoma has not been established. To address the possibility of detecting specific circulating microRNAs years before a B-cell lymphoma is diagnosed, we studied the plasma expression of microRNA first in pre-treatment samples from patients with diffuse large B-cell lymphoma and subsequently in repository samples from blood donors who later developed B-cell lymphomas. In addition, we studied the microRNA expression in the diagnostic lymphoma biopsy and in plasma samples taken after end of treatment. The most strongly induced (miR-326) and suppressed (miR-375) plasma microRNA at diagnosis, when compared with healthy blood donors, were also substantially up- or down-regulated in plasma repository samples taken from several months to up to two years before the blood donors were diagnosed with B-cell lymphoma. Importantly, at these time points the donors had no signs of disease and felt healthy enough to donate blood. In conclusion, this first study of plasma microRNA profiles from apparently healthy individuals, taken several years before B-cell lymphoma diagnosis, suggests that plasma microRNA profiles may be predictive of lymphoma development.

Project description:Exosomes, extracellular vesicles of 30-150 nm, are released by normal and tumor cells. These nanovesicles play a major role in cell communication and can be found in biological fluids as carriers of biomarkers. Tumor exosomes can inhibit immune responses, mediate drug resistance and transform mesenchymal stem cells. In contrast to healthy donors, cancer patients’ plasma contain higher levels of exosomes. Cutaneous melanoma is a very aggressive cancer whose incidence has rapidly increased worldwide and the prognosis is generally poor, given the propensity of melanoma cells to spread to distant sites while evading immune system control.We investigated potential differences between plasma exosomes derived from healthy donors (HD) and melanoma patients at 0-I, II, and III-IV stages of disease.

Project description:Elite controllers maintain HIV-1 viral loads below the limit of detection. The mechanisms responsible for this phenomenon are poorly understood. As microRNAs (miRNAs) are regulators of gene expression and some of them modulate HIV infection, we have studied the miRNA profile in plasma from HIV elite controllers and chronically infected individuals and compared against healthy donors. Several miRNAs correlate with CD4+ T cell count or with the known time of infection. No significant differences were observed between elite controllers and healthy donors; however, 16 miRNAs were different in the plasma of chronic infected versus healthy donors. In addition, levels of hsa-miR-29b-3p, hsa-miR-33a-5p and hsa-miR-146a-5p were higher in plasma from elite controllers than chronic infected and hsa-miR-29b-3p and hsa-miR-33a-5p overexpression significantly reduced the viral production in MT2 cells. Therefore, levels of circulating miRNAs might be of diagnostic and/or prognostic value for HIV infection. Additionally, hsa-miR-29b-3p and miR-33a-5p may be used in therapeutic strategies. An exploratory cross-sectional study of microRNA levels in EDTA plasma samples. Plasma samples were obtained from 24 subjects and were classified in 3 groups, 9 Elite Controllers (defined as individuals with plasma viral load (PVL) < 50 copies/ml, CD4 count >350/ml), 9 chronic HIV patients (CH) under anti-retroviral treatment and 6 healthy HIV negative donors (HD). Project description:The pathogenic determinants of chronic periodontitis leading to extra-oral comorbidity are unclear. This study was conducted to investigate the main determinants that correlate chronic periodontitis and systemic diseases via periodontitis-evoked molecular changes in immune cells resolved by therapy. Single-cell RNA sequencing using peripheral blood mononuclear cells was performed to compare the molecular and biological differences in immune cells of 4 pre- and 4 post-treatment patients and 4 healthy donors. Periodontal treatment was one-day full-arch scaling and root planning for full mouth disinfection. Altered expression of genes involved in the inflammatory response in the patients was compared with that in healthy donors following therapeutic intervention. The common representative genes across diverse cell types were investigated; those genes were associated with periodontitis-pathogenic bacterial systemic diseases. These important factors correlating chronic periodontitis and systemic inflammatory diseases at the single-cell level may serve as therapeutic targets.

Project description:The wide array of molecules carried by plasma regulates critical immune functions and constitutes valuable biomarkers and therapeutic targets. In recent years the introduction of “systems approaches” has provided investigators with powerful means for assessing immune responses in patient samples on a global scale. However, while the use of genome-wide profiling technologies has become widespread, measuring the plasma proteome still presents considerable challenges. An alternative approach that consists in measuring transcriptome responses in reporter cells exposed in vitro to patient plasma has been successfully employed in a limited number of studies. Here we devised such a “Transcriptomic Reporter Assay” system to assess the immunogenicity of plasma from septic patients and evaluate its potential for biomarker discovery. Sepsis is a common, severe systemic infectious process for which physicians still lack efficient diagnostic or prognostic tools. Of the three different cell reporter systems tested, neutrophils were identified as the most capable “plasma sensor”. Compared to peripheral blood mononuclear cells and dendritic cell preparations neutrophils were best able to discriminate between plasma from septic and control subjects and responded by upregulating a robust immune transcriptional program. Additionally, the amplitude of the neutrophil transcriptomic response was shown to be associated with disease severity in two additional sets of patients. Overall, our results demonstrate both the suitability and potential clinical relevance of a neutrophil reporter assay for assessing immunopathogenic processes in a complex and severe condition such as sepsis. Transcriptional profiles were acquired using Illumina HumanHT12 V4 BeadChips.
