Renal medullary genes in salt-sensitive hypertension

Project description:Substitution of chromosome 13 from Brown Norway BN/SsNHsd/Mcw (BN/Mcw) rats into the Dahl salt-sensitive SS/JrHsd/Mcw (SS/Mcw) rats resulted in substantial reduction of blood pressure salt sensitivity in this consomic rat strain designated SSBN13. In the present study, we attempted to identify genes associated with salt-sensitive hypertension by utilizing a custom, known-gene cDNA microarray to compare the mRNA expression profiles in the renal medulla (a tissue playing a pivotal role in long-term blood pressure regulation) of SS/Mcw and SSBN13 rats on either low-salt (0.4% NaCl) or high-salt (4% NaCl, 2 wk) diets. Gene expression profiling of hypertensive and hypotensive mice

Project description:Hypertension is a condition with major cardiovascular and renal complications, affecting nearly a billion patients worldwide. Few validated gene targets are available for pharmacological intervention, so there is a need to identify new biological pathways regulating blood pressure and containing novel targets for treatment. The genetically hypertensive “blood pressure high” (BPH), normotensive “blood pressure normal” (BPN), and hypotensive "blood pressure low" (BPL) inbred mouse strains are an ideal system to study differences in gene expression patterns that may represent such biological pathways. We profiled gene expression in liver, heart, kidney, and aorta from BPH, BPN, and BPL mice and determined which biological processes are enriched in observed organ-specific gene signatures. As a result, we identified multiple biological pathways linked to blood pressure phenotype that could serve as a source of candidate genes causal for hypertension. In order to distinguish causal genes from responsive genes in the kidney gene signature we integrated phenotype associated genes into Genetic Bayesian networks, identifying several novel candidate genes causal for hypertension. The integration of data from gene expression profiling and genetics networks is a valuable approach to identify novel potential targets for the pharmacological treatment of hypertension.

Transcription profiling of rat kidney treated with adrenocorticotropic hormone (ACTH, 0.2mg/kg) or dexamethasone (DEX, 0.02mg/kg) vs. controls

Project description:The left kidneys of 8 Sprague-Dawley rats were removed and the rats were left for 2 weeks to recover. After 4 additional days (during which blood pressure measurements were obtained), they were injected daily subcut. with either saline (1ml/kg), adrenocorticotropic hormone (ACTH, 0.2mg/kg) or dexamethasone (DEX, 0.02mg/kg) for 8 days before the right kidney was removed. ACTH and DEX treatment caused hypertension in the rats as illustrated by increased systolic blood pressure. The gene expression patterns of the kidneys were analysed using Affymetrix arrays. Experiment Overall Design: The left kidney of each rat acts as a control for the treated right kidney. Two rats each were treated with either saline, ACTH or DEX.

Lack of xanthine dehydrogenase leads to a remarkable renal decline in a novel hypouricemic rat model

Project description:The SSXdh-/- rat (SS-Xdhem1Mcwi, Rat Genome Database ID:14398479), created on the Dahl SS background, resulted in severe hypouricemia in the tissues. With the Xdh project, the overall hypothesis is that the Xdh gene ablation will result in kidney damage and perturbation of blood pressure control. We were interested in finding out if the gene ablation alternates the kidney development process. The deletion of Xdh protein produced dramatic changes in proteins related to the salvage pathway. The novel Xdh knockout rat model exhibited profound hypouricemia, which increased oxidative stress and crystal formation, leading to inflammation, renal cell proliferation, severe kidney damage, and a subsequent decline in renal function.

Molecular mechanisms of target organ damage in youth with primary hypertension

Project description:High blood pressure or Primary Hypertension is associated with target organ damage (TOD) in both adults and youth, where TOD predicts the adverse cardiovscular (CV) events. The design and methods of the SHIP AHOY (Study of High Blood Pressure in Pediatrics, Adult Hypertension Onset in Youth) project have been discussed elsewhere (Mendizabal B, et al., 2018, Hypertension. 2018 Sep;72(3):625-631.). Briefly, SHIP AHOY study participants with normal (Cut off: SBP <80th%), and high- systolic blood pressure (Cut off: SBP >90th%), were stratified as with or without TOD, like Low LVMI (Left ventricular mass index) (Cut off: <38.6 gm/m2.7), and High LVMI (Cut off: >38.6 gm/m2.7), were selected in the present study cohort. Blood samples (for PBNCs and serum) were collected to investigate genetic, epigenetic, and proteomics changes that influence the development of hypertensive TOD in youth. We excluded youth with symptomatic severe hypertension, on antihypertensive or lipid-lowering medication in the past 6 months, with diabetes mellitus (type 1 or 2), kidney disease, or other chronic medical conditions. All study participants and their parents provided written informed consent and assent according to local investigational review board requirements. The mechanisms involved in blood pressure mediated TOD are limitedly understood in onset of TOD. To understand this, using RNA sequencing, we first identified candidate differentially expressed genes in circulating PBMCs of study participants. Next, through miRNA and whole genome bisulphite sequencing of circulating PBMCs we determined the epigenetic regulators of TOD. Also, using LCMS, serum proteomics, the key circulatory protein involved in advancing CV- TOD events was identified. In conclusion, multiomics approach was utilized to define the circulatory regulators involved in BP mediated CV-TOD.
