A MKP-MAPK protein phosphorylation cascade controls vascular disease resistance in plants

Project description:Vascular plant diseases, such as rice bacterial blight caused by Xanthomonas oryzae pv. oryzae (Xoo) and crucifer black rot caused by Xanthomonas campestris pv. campestris (Xcc), cause huge yield loss of crops worldwide. However, how plants operate vascular defense against these obligate pathogens remains elusive. In this study, we used both Arabidopsis and rice pathosystems to address the long-standing question. We found that the loss of function mutation of Arabidopsis mitogen-activated protein kinase (MAPK) phosphatase 1 (MKP1) lost the non-host resistance to Xoo and supported Xoo to grow in the leaf veins, which also enhanced susceptibility to Xcc. MKP1 regulates the MPK3-mediated phosphorylation of the transcription factor MYB4 that functions in vascular lignification. Importantly, the MKP-MAPK cascade-mediated lignin biosynthesis is also conserved in rice through regulating OsMYB102 and OsMYB108, which control rice vascular resistance to adapted Xoo. Interestingly, the Arabidopsis and rice mutants enhanced resistance to the mesophyll cell pathogens most likely through upregulating salicylic acid biosynthesis, Pseudomonas syringae (P. syringae) and Xanthomonas oryzae pv. oryzicole (Xoc), respectively; strongly suggesting that this immune mechanism is likely specific to the obligate vascular pathogens. Therefore, our study uncovers a previously unrecognized vascular-specific and lignin-based immune mechanism, shedingshedding new sight on tissue-specific immunity in plants, as well as providing a practical approach for improvement of disease resistance against vascular pathogens in crops

Xanthomonas oryzae pv. oryzae (Indian Isolate): Wild type vs. rpfF mutant

Project description:Transcription profiling of the DSF regulon in Xanthomonas oryzae pv. oryzae (Xoo) using wild type and the rpfF mutant. Cell-cell signaling mediated by the quorum sensing molecule known as Diffusible Signaling factor (DSF) is required for virulence of Xanthomonas group of plant pathogens. DSF in different Xanthomonas and the closely related plant pathogen Xylella fastidiosa regulates diverse traits in a strain specific manner. The transcriptional profiling performed in this study is to elucidate the traits regulated by DSF from the Indian isolate of Xanthomonas oryzae pv. oryzae, which exhibits traits very different from other Xanthomonas group of plant pathogen. In this study, transcription analysis was done between a wild type Xanthomonas oryzae pv. oryzae strain and an isogenic strain that has a mutation in the DSF biosynthetic gene rpfF.

Project description:Common Bacterial Blight (CBB) is a major threat to bean crops caused by Xanthomonas citri pv. fuscans (Xcf). The pathogenicity of Xcf is known to be dependent upon a functional Type 3 Secretion System (T3SS), that allows the injection of numerous Type III Effectors (T3Es) into plant cells. We generated a transcriptomic dataset to compare the response of susceptible and resistant cultivars of Phaseolus vulgaris to the inoculation of the virulent strain Xcf CFBP4885 or its avirulent T3SS-defective hrcV mutant (CFBP13802). This dataset is a valuable resource to investigate the role of T3Es in subverting the cellular functions of bean.

A transcriptome study of DSF-mediated quorum sensing in Xanthomonas axonopodis pv. citri

Project description:Quorum sensing (QS) in Xanthomonas axonopodis pv. citri, the causal agent of citrus canker, is mediated by a diffusible signal factor (DSF). QS is required for the full virulence of X. axonopodis pv. citri in planta. Mutations in rpfF, rpfC and rpfG, the core genes of QS, decreased the production of extracellular proteases and bacterial motility. Comparison of the transcriptomes of QS mutants with that of the wild type stain revealed that QS temporally regulates the expression of a large set of genes, including genes involved in chemotaxis and flagellar biosynthesis, genes related to metabolism, genes encoding virulence traits such as type II secretion system substates, type III secretion system and effectors. Cross talk between the QS regulon and the HrpG regulon has also been identified by 62 common genes shared by both regulons. The temporal regulation of the QS regulon and cross talk with the HrpG regulon suggest the important role of QS in citrus canker infection, including attachment, invasion and growth in host apoplast. Three mutants and two time-points experiment. Four biological and dye-swap replicates of each strain per each time-point were used. In total, there were two datasets of the rpfF mutant: 1) rpfF mutant vs. wild type strain at 11 h, and 2) rpfF mutant vs. wild type strain at 25 h; two datasets of the rpfC mutant: 1) rpfC mutant vs. wild type strain at 11 h, and 2) rpfC mutant vs. wild type strain at 25 h; and two datasets of the rpfG mutant: 1) rpfG mutant vs. wild type strain at 11 h, and 2) rpfG mutant vs. wild type strain at 25 h.
