Low molecular weight polysialic acid prevents lipopolysaccharide-induced inflammatory dopaminergic neurodegeneration in humanized SIGLEC11 transgenic mice

Project description:Here, we investigated the effect of low molecular weight polysialic acid (polySia avDP20) on inflammatory neurodegeneration. SIGLEC11 transgenic and wild type control (WT) mice were treated intraperitoneally daily over four consecutive days with 1 µg/gbw/day LPS in conjunction with 10 μg/gbw polySia avDP20. Repeated LPS injections led to inflammation and neuronal loss in the brains of WT and SIGLEC11 transgenic mice. Transcriptome analysis by RNA-seq of total brain tissue indicated that treatment with polySia avDP20 prevented the LPS-induced upregulation of genes belonging to inflammation, TLR signaling, phagocytosis, and complement-related pathways in SIGLEC11 transgenic but not wild type mice.

Project description:Here, we investigated the role of mononuclear phagocytes associated nicotinamide adenine dinucleotide phosphate oxidase 2 (NOX-2) in inflammatory neurodegeneration. Cybb-deficient mice NOX-2 knock-out (KO) and control wild type (WT) mice were treated intraperitoneally daily over four consecutive days with 1 µg/gbw/day LPS. Repeated LPS injections led to inflammation and neuronal loss in the brains of WT mice, which was attenuated after knockout of NOX-2. Transcriptome analysis by RNA-seq of total brain tissue indicated increased LPS-induced upregulation of genes belonging to the reactive oxygen species (ROS) and reactive nitrogen species (RNS) production, complement and lysosome activation as well as apoptosis and necroptosis in WT compared to NOX-2 KO mice.

Initial effects of BBN treatment on mouse bladder

Project description:Characterizing the initial stages of oncogenic transformation in bladder cancer. Littermate male mice were transferred in single-use plastic cages in order to be treated with the BBN carcinogen. The treatment dose used in this work was 0.05% BBN delivered in drinking water for up to 12 days. Bladder samples were collected at day 1, 4, 8 and 12 from treated mice along with the age-matched controls (non-treated). The drinking water was changed every fourth day. The weights of the mice and BBN water intake were measured every four days and mice were checked regularly for any signs of discomfort and pain.

Repeated systemic challenge with lipopolysaccharides induces dopaminergic neurodegeneration in the brain by activation of the complement-phagosome pathway

Project description:Systemic inflammatory reactions mediated by chronic infections activate microglia in the central nervous system (CNS) and have been postulated to exacerbate neurodegenerative diseases. We now demonstrate in vivo that repeated systemic challenge of mice with bacterial lipopolysaccharides (LPS) maintains an elevated microglial inflammatory response and triggers neurodegeneration. Repeated chronic intraperitoneal application of LPS over four consecutive days induced loss of dopaminergic neurons in the substantia nigra, a process that was accompanied by decreased levels of dopamine in the striatum. In contrast, total cumulative LPS dose given intraperitoneally within a single acute application did not induce a decrease in dopamine levels nor neurodegeneration. Mice that received repeated systemic LPS application showed increased microglial activation, elevated production of proinflammatory cytokines and activation of the classical complement and its associated phagosome pathway in the brain. Loss of dopaminergic neurons induced by repeated systemic LPS application was rescued in complement C3 deficient mice, confirming an involvement of the complement system in neurodegeneration. Thus, our data demonstrate that repeated systemic exposure to bacterial LPS induces a microglial phagosomal inflammatory response, leading to complement-dependent damage of dopaminergic neurons.

Genome-wide analysis of chronic inflammation induced gene expression in livers isolated from either wild type or ApoE-Cyp7a1 transgenic animals.

Project description:Analysis of whole genome expression changes in livers from wild type animals and animals with a liver specific transgenic over expression of Cyp7a1. Mice were given a chronic, repetitive administration of LPS for 7 days. Our prior analysis had indicated that inflammation suppresses Cyp7a1 and that this leads to accumulation of intermediates in the mevalonate biosynthesis pathway. Here, we hypothesized that over expression of Cyp7a1 would not affect the changes in transcriptional state due to chronic administration of LPS. We provide gene expression data which evaluates this question. Here we find that over expression of Cyp7a1 minimally alters the transcriptome of livers in an untreated state, and that it has small effects on the response to chronic LPS. Total RNA isolated from livers of wild type and liver specific Cyp7a1 transgenic animals treated with or without recurrent, daily LPS injections (1.5mg/kg) for 7 days. There are two biological replicates per condition. Samples are a matrix of all conditions reported as FPKMs.

Genome-wide anyalysis of gene expression during prolonged endotoxin tolerance in Aoah -/- mice

Project description:Analysis of gene expression in explanted peritoneal macrophages from Aoah -/- and Aoah +/+ mice treated with LPS 21 days prior to harvest. Explanted peritoneal machrophages were challenged with LPS or control (PBS). The study seeks to characterize global gene expression in the state of prolonged LPS tolerance induced in mice lacking the LPS-inactivating enzyme Aoah. Groups of 9 C57BL/6 Aoah+/+ or Aoah-/- mice were given i.p. injections of 10 µg E. coli LPS/mouse. Twenty-one days later (when Aoah-/- mice remain tolerant and Aoah+/+ mice have recovered), peritoneal macrophages were harvested and the yields from three mice were pooled to form 3 samples per group (i.e., three samples of Aoah+/+ and Aoah-/- mice, with each sample comprised of peritoneal macrophages from 3 mice). Next day, cells were challenged with LPS or PBS and whole RNA was isolated 2 hours later and used for microarray experiments.
