Project description:Hematopoietic stem cells (HSCs) constitute a rare cell population in bone-marrow and are capable of live-long self-renewal and production of all mature blood cell types. Cell differentiation processes are governed by epigenetic mechanisms whose study during early differentiation steps will provide insights into stem cell function and differentiation. We performed whole-genome bisulfite sequencing on HSCs and their immediate progeny, namely three different multipotent progenitor subpopulations (MPP1, MPP2, and MPP). Whole-genome bisulfite sequencing of hematopoietic stem cells (HSCs) and 3 different multipotent progenitor subpopulations (MPP). Three independent biological replicates each were analyzed.

Proteomics profiling of human CD34+ cells from MDS bone marrow and healthy control cells

Project description:Myelodysplastic syndromes (MDS) are a heterogeneous group of hematopoietic disorders characterized by ineffective blood cell production and a high risk of progression to acute myeloid leukemia (AML). CD34+ hematopoietic stem and progenitor cells (HSPCs) play a critical role in the pathophysiology of MDS, yet the proteomic changes underlying the disease remain poorly characterized. This project focuses on performing comprehensive quantitative proteomic profiling of CD34+ cells isolated from the bone marrow of MDS patients and healthy controls. By leveraging advanced mass spectrometry and quantitative proteomics techniques, we aim to identify unbiased differences in protein expression and pathways between diseased and healthy cells. These findings will contribute to a deeper understanding of the molecular mechanisms driving MDS and may reveal potential biomarkers or therapeutic targets.

Clonal Evolution of Pre-Leukemic HSC Precedes Human AML

Project description:<p>Acute myeloid leukemia is an aggressive clonal malignancy of the bone marrow that is the direct result of sequential acquisition of mutations in a single lineage of cells. In this study, we investigate a model in which this mutational acquisition occurs serially in long-lived self-renewing hematopoietic stem cells eventually resulting in frank acute myeloid leukemia. Coding mutations in multiple AML patients were identified using exome sequencing followed by sanger sequencing validation. The level of these mutations was then assessed in residual hematopoietic stem cells from each patient using targeted deep sequencing. These population-level estimates of mutant allele burden were then validated in single cell assays targeted to the identified mutations. This allowed for determination of the order of acquisition of the mutations that preceded the development of the leukemia. The results of this study identify pre-leukemic hematopoietic stem cell clones that could contribute to patient relapse and outcome.</p>
