Differential gene expression profiles of side population and non-side population cells of malignant pleural mesothelioma

Project description:Malignant pleural mesothelioma (MPM), which is associated with occupational asbestos exposure, is a deadly disease with no effective treatments due mainly to its high resistance to anti-cancer drugs. The molecular mechanisms responsible for its chemotherapeutic resistance are complicated and undefined. However, the presence of side population cells (SP cells) in tumors is a well-accepted explanation for their anti-cancer drug resistance. To identify SP cell-specific gene expression signature, microarray technique has been employed. Our data show differential gene expression profiles between SP and non-SP cells of H2714 mesothelioma cells. SP cells over-expressed genes associated with cancer stem cell (CSC) and drug resistance: DUSP6, SPRY2 and IL6, as well as multi-pathways, including the cancer stem cell-associated pathways Notch and c-Kit. Therefore, we believe that targeting CSC-specific genes and pathways in SP cells may hold the key to the discovery of effective treatments for reversing chemotherapeutic resistance to MPM treatment. 4 samples

Establishment and characterization of fluorouracil and gemcitabine resistant cholangiocarcinoma cell lines

Project description:Resistance to commonly used chemotherapeutic drugs is a major problem of cholangiocarcinoma (CCA) therapy, therefore more effective treatment strategy is urgently needed. Drug resistant cell lines have been used for elucidating drug resistance mechanisms in various cancers leading to the discovery of novel treatments. Here in, we established a 5-Fluorouracil and Gemcitabine resistant CCA cell lines. The resistant sublines were selected based on constant exposure of KKU-213B parental cells to 5-Fluorouracil and gemcitabine in a stepwise dose incremental strategy starting from below IC25 concentration). The cells that grown normally in the presence of 5-FU 7 and Gem 3 μM were used for subsequence experiments and weredesignated as KKU-213B-FR and KKU-213B-GR. The resultant cell lines were confirmed to be resistant to chemotherapeutic drug and also increased cell migration and invasion activity. In addition, the quantitative LC-MS/MS were utilized for the analysis of proteins involved in drug resistance mechanisms.

The landscape of therapeutic vulnerabilities in EGFR inhibitor osimertinib drug tolerant persister cells [ChIP-seq]

Project description:Third-generation EGFR tyrosine kinase inhibitors (EGFR-TKIs), including osimertinib, an irreversible EGFR-TKI, are important treatments for non-small cell lung cancer with EGFR-TKI sensitizing or EGFR T790M resistance mutations. Whilst patients treated with osimertinib show clinical benefit, disease progression and drug resistance are common. Emergence of de novo acquired resistance from a drug tolerant persister (DTP) cell population is one mechanism proposed to explain progression on osimertinib and other targeted cancer therapies. Here we profiled osimertinib DTPs using RNA-seq, ChIP-seq, and ATAC-seq to characterize the features of these cells and performed drug screens to identify therapeutic opportunities.
