Comparison of young (8 weeks) and aged (110 weeks) CD11C+ splenic dendritic cells from C57BL/6 mice

Project description:Total RNA was isolated from FACS purified CD11c+ splenic DC populations isolated from young (8 weeks old, n=3) and aged (110 weeks old, n=3) mice, that had been injected with Poly I:C for 4h previously. Microarray analysis was performed with the Affymetrix GeneChip Mouse Genome 430 2.0 Arrays. The arrays were hybridized, washed and scanned according to the standard Affymetrix protocol. The gene chip tests were performed by professional staff of Shanghai Biochip Company, PRC. Gene expression profiles and functional gene set enrichment analysis demonstrate that DC isolated aged mice are impaired in their ability to activate Natural Killer (NK) cells. The study is submitted for publication in June 2014.

Aging-triggered iron-rich insoluble protein aggregates in the spleen originate chiefly from red pulp macrophages.

Project description:Both iron homeostasis and erythropoiesis are known to be affected by aging. Iron needs in mammals are met primarily by iron recycling from senescent red blood cells (RBCs), a task chiefly accomplished by red pulp macrophages (RPMs) in the spleen. Given that RPMs continuously process iron, their cellular functions might be susceptible to age-dependent decline, a possibility that has been unexplored to date. In our project, we identified a formation of undegradable iron- and heme-rich extracellular aggregates in the spleens of 10-11-month-old female mice. To better understand the origin of these aggregates, here, we performed: i) protein identification and intrasample quantification (iBAQ) of proteins of magnetically-isolated red pulp macrophages from spleens of two female 8-weeks-old C57BL/6J (maintained on a standard diet) and ii) label-free quantification of proteins of the splenic protein aggregates formed in the mouse spleen 24 hours after intraperitoneal iron dextran injection, using dextran-injected mice as a control. Two 8-week-old C57BL/6J mice per group were analyzed. This dataset is related to the project PXD032900, which describes quanytitative analysis of proteins in aggregates magnetically isolated from spleen of aged (standard or iron-reduced diet) and young mice (standard diet). The dataset was obtained with the support of the National Science Centre Sonata Bis grant (UMO-2020/38/E/NZ4/00511).

Four samples from four mice were sorted and analyzed where in each littermates pair consisted of a control and Runx3 conditional KO. Mice lacking Runx3 specifically in the DC compartment were produced by crossing Runx3fl/fl mice onto CD11c-Cre mice. This mating scheme generated Runx3fl/fl/CD11c:Cre (CD11c-DC-Runx3Δ) mice.

Influence of type I Interferons on function of splenic conventional dendritic cells.

Project description:Type I Interferons encompasses a large family of closely related cytokines comprising of at least 13 IFN-α isotypes and single IFN-β. Both IFN-α and IFN-β exert their activity through a common receptor IFNAR. Type I Interferons have broad regulatory effects and various subtypes of dendritic cells are influenced by this cytokines. In our study we asked question whether the low, constitutive levels of type I Interferons produced under steady state conditions are important for proper function of splenic conventional dendritic cells. In this approach we sorted out two populations (CD8α+ and CD8α-) of splenic dendritic cells (DCs) from untreated WT, IFN-β-/- and IFNAR-/- C57Bl/6 mice. All mice were between 8-10 weeks old. Further we isolated RNA and performed microarray analysis. Each DCs population was repeated twice.

Age-associated mouse brown adipose tissue mitochondrial proteome

Project description:We investigated the age-associated thermogenesis activation in brown fat mitochondria. We administered the young mice (10 weeks old) and old mice (76 weeks old) with a selective β3-adrenergic receptor agonist (β3-AR agonist, CL316,243) at a high dose (1 mg kg-1) for five days at 30˚C to activate thermogenesis. We isolated mitochondria from brown fat for proteome profiling. We applied 250 ng peptides for LC/MS/MS analysis using label-free quantification coupled match-between-run approach.
