Transcriptomic profiling of chicken lung epithelial cells during low pathogenic avian influenza infection

Project description:The aim of the study was to assess the effects of low pathogenic avian A/turkey/Massachusetts/3740/1965 [H6N2] influenza virus on a novel chicken lung epithelial cell line CLEC213. CLEC213 cells were mock-infected (4 samples) or infected with H6N2 virus (4 samples) at a multiplicity of infection (MOI) 1. Total RNA was collected 24 hours post-infection. To reduce potential experimental biases, RNA samples were collected from 4 independent experiments.

Cynomolgus macaque model of H7N9 influenza infection

Project description:TheÂ objectiveÂ of this study is to characterize the response to newly emerged, highly pathogenic H7N9 influenza virus isolated from human patients in 2013 in China. This study examines the pathogenesis of H7N9 influenza in cynomolgus macaques.Â The study compares lung lesions to adjacent right lower lobe lung tissue in animals necropsied at days 3 and 6 post-infection (n=4 animals/timepoint).Â 3-4 lesions from each animal were collected and equal amounts of pooled RNA from lesions from individual animals at each time point were used for microarray. 8 cynomolgus macaques were infected via oral, intraocular, intranasal, and intratracheal administration of a combined total of 7x10^6 TCID50. Lungs, lung lesions, and trachea samples were collected from serial sacrifices of 4 animals each at day 3 and day 6. Infection produced a moderate-severe, self-limiting respiratory infection, and was not lethal. We performed microarray analysis (using Agilent Rhesus arrays) on all lungs, lung lesions, and trachea collected for the study.

Two chickens were infected with each 10^6EID50/ head virus intranasally, and their lung was collected from infected chicken at 24 hours after infection.

Expression data of lungs of mice infected with two highly pathogenic avian influenza viruses

Project description:We isolated two highly pathogenic H5N1 avian influenza viruses (AIVs) (CK10 and GS10) with similar genetic background but greatly differ in pathogencity in mice. CK10 is highly pathogenic in mice, whereas GS10 is nonpathogenic. However, the host mechanism of this differecne in pathogenicity is unclear. We used microarray analysis to evaluate the global transcriptional response in the lung of mice infected with CK10 or GS10. Six-week-old female BALB/c mice were inoculated with CK10 or GS10 or mock-infected. At day 1 post infection, lungs were isolated from three mice per group for RNA extraction and hybridization on Agilent microarrays. Samples were named as follows: CK10 (CK10_1_NS,CK10_2_NS,CK10_3_NS) GS10 (GS10_1_NS,GS10_2_NS, GS10_3_NS) control (C1_NS, C2_NS, C3_NS).
