Glioblastoma-derived extracellular vesicles facilitate transformation of astrocytes via reprogramming oncogenic metabolism

Project description:Background: Glioblastoma (GBM) may arise from brain astrocytes through a multistep process that occurs by the temporal accumulation of genetic mutations. Here, we explore whether GBM-derived extracellular vesicles (EVs) may facilitate neoplastic transformation and malignant growth of astrocytes. Methods: We utilized conditioned medium (CM) of glioma cell and stem cell, its sequential filtration, diverse cell-based assays, RNA sequencing, and metabolic assays to compare the effects of EV-containing and EV-depleted CM on the transformation of human and mouse astrocytes in vitro and tumor growth in vivo. Results: GBM EVs facilitated the neoplastic growth of astrocytes pre-transformed by oncogenic mutations or viruses but were unable to transform normal human and mouse astrocytes. GBM EVs induced proliferation, self-renewal, and colony formation of pre-transformed astrocytes, and enhanced astrocytoma growth in a mouse allograft model. Analysis of extracellular RNAs (exRNAs) in GBM identified multiple full-length mRNAs encoding ribosomal proteins, oxidative phosphorylation, and glycolytic factors. GBM EVs appear to reprogram astrocyte metabolism by inducing a shift in gene expression that may be partly associated with EV-mediated mRNA transfer from glioma cells. Conclusions: Our study suggests a novel EV/exRNA-mediated mechanism contributing to astrocyte transformation via metabolic reprograming and implicates horizontal mRNA transfer in this process.

Transcription profiling of mouse C57Bl and nude strain xenografts from adherent and stem cells

Project description:Astrocytes may give raise to glioma, the most frequent primitive CNS tumours. TGFa, an EGF family member, is trophic for both astrocytes and gliomas cells and is frequently over-expressed in the early stages of glioma progression. Although its sole deregulation is insufficient to lead to cancerous transformation of astrocytes, it triggers their conversion into neural progenitor-like cells. We determined whether astrocytes converted into neural progenitor-like cells are more sensitive than their mature counterparts to cancerous transformation using gamma irradiation. Control and TGFa-treated astrocytes had identical cytogenomic profiles. Irradiation did not modify the lifespan of mouse astrocytes cultivated in serum-free medium. On the opposite TGFa-treated astrocytes were immortalized upon irradiation. Their ability to form colonies in methylcellulose medium, their cytogenomic anomalies, and the formation of high-grade glioma-like tumours after grafting into mice CNS testified to their cancerous transformation. Sham-irradiated TGFa-treated astrocytes displayed no evidence of transformation. These results demonstrate that instability of the mature astrocyte phenotype due to a single change in their environment is sufficient to sensitize them to an oncogenic stress. They allow proposing that TGFa does not only favour growth and survival of established gliomas but participates also to the earliest stages of their genesis. Rapid reprogramming of primary human astrocytes into potent tumor initiating cells with defined genetic factors

Project description:Cancer stem cells play key roles in human malignancies. However, their cellular and molecular origins are still poorly understood. Here we report the successful generation of cancer stem cells from primary human astrocytes by use of defined genetic factors. Combined transduction of four factors âmyc, Oct-4, p53DD, and rasâ induced efficient transformation of primary human astrocytes into malignant cells with powerful tumor-initiating capabilities. Transplantation of 100 of the cells into nude mice is sufficient for tumor formation. The cells also showed unlimited self-renewal ability with robust telomerase activities. In addition, they expressed typical glioma stem cell markers such as CD133, CD15, and CD90. Moreover, transformed cells could form spheres in culture and differentiate into neuron-astrocyte-, and oligodendrocyte-like cells. They also displayed significant resistance to temozolomide. These induced cancer stem cells (iCSCs) should be useful for both tumor biology studies and therapeutics development. This record contains data from microarray analysis of normal human astrocytes, two transduced cell lines (OCT4+3G and OCT4+6G), a patient-derived glioma stem cell line (ALPS1459) and a glioblastoma cell line (U87MG).

Gene expression analysis of astrocytes differentiated from iPS cells generated from the fibroblasts of PSEN1deltaE9 mutant AD and their isogenic control iPS lines

Project description:Mutations in presenilin 1 (PSEN1) cause a familiar form of Alzheimer's disease (AD). We have obtained skin biopsies from two individuals carrying PSEN1 exon9 deletion and reprogrammed skin fibroblasts into induced pluripotent stem cells (iPSCs). To get controls, we have corrected PSEN1 exon9 deletion by CRISPR/Cas9 technique. Since astrocytes play role in AD pathogenesis, we further differentiated iPSCs into astrocytes. We ran RNA sequencing analysis to compare our iPSC-derived astrocytes with previously published data on human astrocytes and to identify genes and pathways affected by PSEN1 exon 9 deletion.

Context-dependent Regulation of Notch Signaling in Glial Development and Tumorigenesis [Bulk RNA-seq]

Project description:In the mammalian brain, Notch signaling maintains the cortical stem cell pool and regulates the glial cell fate choice and differentiation. However, the function of Notch in regulating glial development and its involvement in tumorigenesis have not been well understood. Here, we show that Notch inactivation by genetic deletion of Rbpj in stem cells decreases astrocytes but increases oligodendrocytes with altered internal states. Interestingly, inhibiting Notch in glial progenitors does not affect cell generation but instead accelerates the growth of Notch-deprived oligodendrocyte progenitor cells (OPCs), and OPC-related glioma. We also identified a crosstalk between oligodendrocytes and astrocytes, with premyelinating oligodendrocytes secreting Bmp4, which is repressed by Notch, to upregulate GFAP expression in adjacent astrocytes. Moreover, Notch inactivation in stem cells causes a glioma subtype shift from astroglia-associated to OPC-correlated patterns, and vice versa. Our study reveals Notch's context-dependent function, promoting astrocytes and astroglia-associated glioma in stem cells and repressing OPCs and related glioma in glial progenitors.
