Functional targeting of PTTG1 in pericytes restores vascular integrity in diabetic retina

Project description:Pericyte loss is an early and critical event in the pathogenesis of diabetic retinopathy (DR), yet the molecular mechanisms underlying pericyte dysfunction remain incompletely understood. Using single-cell RNA sequencing, we generated a retinal cellular overview comprising 37,982 cells from diabetic and non-diabetic mice. We identified a previously unrecognized pericyte subpopulation defined by high expression of pituitary tumor transforming gene 1 (PTTG1), which was enriched in diabetic retina. Functional studies demonstrated that CRISPR/Cas9- or siRNA-mediated silencing of PTTG1 restored pericyte stability and barrier-supporting function under high glucose stress. In vivo, PTTG1 silencing via viral or pericyte-specific AAV delivery improved retinal vascular integrity and reduced retinal vascular dysfunction in diabetic mice. Integrated transcriptomic and metabolomic profiling revealed that PTTG1 silencing reprogrammed metabolism, modulating glycolytic flux and attenuating oxidative stress.

Project description:This study investigates transcriptomic changes in human coronary artery endothelial cells (HCAEC) following siRNA-mediated gene silencing. Cells were transfected with either scrambled control siRNA or siRNA targeting VKORC1 or VKORC1L1. Total RNA was extracted and subjected to bulk RNA sequencing to assess differential gene expression profiles associated with loss of VKORC1 or VKORC1L1 function. The dataset enables analysis of gene expression changes related to vitamin K metabolism and downstream signaling pathways in endothelial cells.

Nuclear pore complexes mediate subtelomeric gene silencing by regulating PCNA levels on chro atin

Project description:The nuclear pore complex (NPC) physically interacts with chromatin and regulates gene expression. The Saccharomyces cerevisiae inner ring nucleoporin Nup170 has been implicated in chromatin organization and the maintenance of gene silencing in subtelomeric regions. To gain insight into how Nup170 regulates this process, we used protein-protein interactions, genetic interactions, and transcriptome correlation analyses to identify the Ctf18-RFC complex, an alternative proliferating cell nuclear antigen (PCNA) loader, as a facilitator of the gene regulatory functions of Nup170. The Ctf18-RFC complex is recruited to a subpopulation of NPCs that lack the nuclear basket proteins Mlp1 and Mlp2. In the absence of Nup170, PCNA levels on DNA are reduced, resulting in the loss of silencing of subtelomeric genes. Increasing PCNA levels on DNA by removing Elg1, which is required for PCNA unloading, rescues subtelomeric silencing defects in nup170Δ. The NPC therefore mediates subtelomeric gene silencing by regulating PCNA levels on DNA.

GnRH Agonist-Induced Ovarian Suppression and Ovarian Steroids in PMDD and Controls

Project description:<p>In this study, lymphoblastoid cell line cultures (LCLs) from women with premenstrual dysphoric disorder (PMDD) and asymptomatic controls were compared via whole-transcriptome sequencing (RNA-seq) during untreated (ovarian steroid-free) conditions and after estradiol and progesterone treatment. In untreated LCLs, pathway analysis of the LCL transcriptome revealed, among others, over-expression of ESC/E(Z) complex genes (an ovarian steroid-regulated gene silencing complex) with significant overexpression of MTF2, PHF19, and SIRT1 (P<0.05) in cells from women with PMDD compared with controls. Finally, mRNA expression of several ESC/E(Z) complex genes were increased by progesterone in controls only, and decreased by estradiol in PMDD LCLs. </p>

To elucidate the signaling pathways that were significantly altered following the silencing of FGFRL1 expression, we performed global gene profiling experiments of the OVCAR8 and ES2 cells after knockdown of FGFRL1 using siRNA. We conducted pathway analysis with the differentially expressed genes using R in two OC cells.
