Early transcriptional changes in Medicago truncatula upon Nod factor treatment

Project description:This experiment was part of a larger study to elucidate the early effects of Nod factor treatment on the Medicago truncatula phosphoproteome, transcriptome and metabolome in wildtype and two mutants. In this experiment, RNA-Seq was used to measure transcriptional changes in plant roots treated +/- Nod factors for 1 hr. All plants were treated for one hour by replacement of the medium with modified Fahraeus medium with and without 10â8 molar Nod factors obtained from Sinorhizobium meliloti strain Rm1021 pRmE43 (pTE3:nodD1). Seedlings were harvested after one hour of treatment. RNA was extracted and sequenced from three biological replicates of each treatment condition in wildtype as well as mutants in nfp and dmi3, two genes known to be involved in the response pathway.

Expression data of Medicago truncatula Jemalong A17 roots treated with auxin transport inhibitors

Project description:Rhizobium and allied bacteria form symbiotic nitrogen-fixing nodules on legume roots. Plant hormones appear to play a role in nodule formation. We treated Medicago truncatula roots with auxin transport inhibitors (ATIs) N-(1-naphthyl)phthalamic acid (NPA) and 2,3,5-triiodobenzoic acid (TIBA) to induce the formation of pseudonodules. We compared the transcriptional responses of M. truncatula roots treated with ATIs to roots inoculated with Sinorhizobium meliloti. The transcriptional response of M. truncatula roots 1 and 7 days after ATI treatment were opposite to roots treated with S. meliloti.

Overexpression of MtABI3 in transgenic Medicago truncatula roots

Project description:ABI3 is a B3-domain transcription factor that acts as a master regulator of seed maturation. To identify genes that are regulated by this transcription factor in the model legume Medicago truncatula, Medicago hairy roots were generated using Agrobacterium rhizogenes transformed with the genomic sequence of the ABI3 gene of Medicago. Using the Medicago NimbleGen chip, a transciptomic analysis was performed to identify differentially expressed genes compared to the GUS expressed control. Two-condition experiment, GUS versus genomic. Biological replicates: 3 control (35S::GUS), 3 treatment (35S::Genomic ABI3 sequence), independently grown and harvested. One replicate per array.

A transcript profile of Medicago truncatula 2HA tissue culture with auxin treatment

Project description:Leaf explants of the superembryogenic Medicago truncatula line 2HA were treated with auxin (1-naphthaleneacetic acid) for one week to induce the formation of roots (Imin et al J Exp Bot 58:439-451). Gene expression in the leaves and the NAA treated tissue cultures was compared to identify transcripts expressed during the commitment to root formation in tissue culture. We have used the Affymetrix Medicago Genome Array GeneChip to compared gene expression in Medicago truncatula leaves and leaf explants that have been cultured for one week on NAA, to identify genes expressed during the commitment to root formation in tissue culture.
