Project description:Oral cancer is characterized by poor prognosis and low survival rate despite sophisticated surgical and radiotherapeutic modalities. Lymphatic metastasis of oral cancer is a complex process involving multiple post-transcriptional biological processes. Thus, feasible detection of cancer metastasis in plasma could be challenging and beneficial. We recruited patients with oral squamous cell carcinoma (OSCC), one of the common oral cancer types, and checked the lymphatic metastatic status and profiled small RNA in plasma samples.

Transcriptome profiling of CD44highCD24low(CSC) and CD44lowCD24high(Non-CSC) SCC25 cells

Project description:In the present study, we have characterized the putative Cancer Stem Cell population of Oral Squamous Cell Carcinoma by various cellular and molecular assay. Subsequently we performed gene expression profiling of SCC25 cell line with CD44highCD24low(CSC) and CD44lowCD24high(Non-CSC) phenotypes using illumina BeadChip Array. Further, systematic computational analysis was performed to identify CSC-like gene signatures in the oral cancer cells. Differentially expressed genes were subjected to pathway analysis in IPA. The analysis lead to the identification of few relevant signaling pathway implicated in stemness.

Project description:DNA methylation is one of the most studied epigenetic alterations in cancer. Genome-wide DNA methylation profiling was conducted in 6 oral tongue squamous cell carcinomas and matched normal tissues. In the present study, the Illumina Infinium HumanMethylationEPIC BeadChip (EPIC array) was used to characterize the DNA methylation pattern across approximately 850,000 CpG dinucleotide methylation loci using DNA isolated of formalin-fixed and paraffin-embedded tissue sections.
