Project description:The goal of the experiment is to describe the overall transcriptomic alterations and signaling pathways activated by T cells upon transplantation in NSG or NSG-HLA-A2/HHD mice. PBMCs and CD3/CD28 activated T cells serve as negative and positive controls of these pathways activation, respectively. PBMCs from 4 different healthy volunteer donors were used. PBMCs from each donor were either used to sort T cells (by FACS) immediately after Ficoll isolation, or immediately stimulated in vitro or injected into mice.

Project description:Our studies have shown that CD44hiCD62LloPSGL-1loCD4+ T (PSGL-1loCD4+ T) cells play an important role in chronic GVHD pathogenesis (R. Deng et al: Nature Communications 2017). Thus, PSGL-1loCD4+ T cells were sorted from spleen of chronic GVHD BALB/c recipient mice transplanted with spleen cells and T cell-depleted bone marrow (TCD-BM) cells from C57BL/6 donor mice. The control no GVHD BALB/c recipient mice were transplanted with C57BL/6 donor TCD-BM only. 21 days after transplantation, PSGL-1loCD4+ T cells from spleen of chronic GVHD and control mice were sorted and subjected to RNA-seq analysis.

Microarray comparison of NK cell from healthy donors and day 14 post stem cell transplantation

Project description:NK cells were first isolated from PBMCs using EasySepâ¢ Human NK cell enrichment kit (STEMCELL biotechnology). The magnetically isolated NK cells were stained with anti-CD3 FITC, anti-CD56 Pe-Cy7 and LIVE/DEADÂ® red stain for a further enrichment, whereby the CD3 negative and CD56 positive live cells were sorted with MoFloâ¢ cell sorter (Beckman Coulter). The sorted NK cell populations were sent to AROS Applied Biotechnology A/S (Aarhus N, Denmark) to be analysed using GeneChipÂ® Human Transcriptome Array 2.0 (Affymetrix, USA). The transcription profile of NK cells from day 14 post stem cell transplantation was compared with those from healthy donors.

Transcriptome of activated human and mouse MAIT cells

Project description:We sought to describe in detail the consequences of MAIT cell activation using a transcriptomic approach to define the basic transcriptome of a MAIT cell in both humans and mice and to determine how this is modulated by activation. Fresh human peripheral blood cells were obtained from three donors. These were cultured for 6 hours with (‘stimulated’) or without (‘unstimulated’) 10 nM 5-OP-RU, magnetically enriched on MR1-tetramer+ cells, and flow-sorted for RNA sequencing of live CD3+TCR Valpha7.2+ MR1-5-OP-RU tetramer+ MAIT cells, and of unstimulated naïve live CD8+CD45RA+ T cells as a comparator cell type. For the murine samples we included within the same sequencing experiment live pulmonary CD3+45.2+19-MR1-5-OP-RU tetramer+ MAIT cells which were magnetically enriched and flow-sorted from the lungs of mice 7 days after infection with 1x104 CFU L. longbeachae (‘acute’), or at least 12 weeks post infection (‘resolution’) or 7 days after a second intranasal infection with 2x104 CFU L. longbeachae in mice that had recovered from infection 12 weeks previously (‘reinfection’). Live CD3+CD45.2+CD19-CD8+CD44-CD62L+ naïve T cells from uninfected mice were used as a comparator cell type.

ImmPRes - PBMC derived CD4 and CD8 T cells

Project description:Dataset release from the Immunological Proteome Resource (ImmPRes). This dataset contains proteomic data from human CD4 and CD8 T cells isolated from PBMC. T-cells were activated with OKT3 antibody (anti CD3) and IL-2 for three days, then expanded in IL-2 until day 7. Cells were then sorted into live CD4 and CD8 T-cells.
