Project description:Deficiency for the enzyme steroid sulfatase (STS) has been associated with postpartum behavioural phenotypes in mice and humans. Here, we compared whole brain gene expression in new mouse mothers acutely treated (two administrations, 48hrs apart) with the STS inhibitor 667-Coumate (10mg/kg p.o.) or vehicle solution (n=12 per group, 3 biological replicates each comprised of 4 pooled individual hemibrain samples).

Phosphorylation of the transcriptional coregulator alphaNAC by DNA-PKcs regulates osteoblastogenesis

Project description:In order to identify pathways affected by inhibition of the DNA-dependent protein kinase, catalytic subunit in osteoblasts, we performed a whole genome gene expression profiling using the Affymetrix mouse GeneChip system (MoGene-2.0 gene ST arrays). The osteoblastic cell line MC3T3-E1 was grown in osteogenic medium and treated with DMSO (vehicle) or NU7441, an inhibitor of DNA-dependent protein kinase, for 14 days. Six biological samples were used in this experiment, three replicates in the control group and three replicates in the treated group. Isolated RNA was subjected to microarray analysis.

Project description:HeLa cells were treated with myristoylation probes X3 and X10, which incorporate both alkyne and diazirine moieties. Cells were then treated with NMT ihibitor IMP-366 (DDD85464) or DMSO vehicle and UV-irradiated before lysis. The experiment identifies myristoylated proteins through comparison of vehicle and NMT inhibitor treated samples. A number of non-myristoylated proteins were enriched together with myristoylated proteins indicating possible photocrosslinked protein interactors.

Effect of chronic glucocorticoid (GC) treatment on neonatal mouse whole cerebellum and whole lung

Project description:Dexamethasone (1ug/g, Dex) or vehicle (saline) injected into postnatal days 0-7 (P0 to P7) of C57BL6J mouse pups every day. Whole organ were harvested at P7. Biological replicates (n = 4) per condition: whole cerebellum or whole lung, treated with dexamethasone or vehicle. This microarray experiment was performed concurrently with the study described in PMID:19164857. Project description:We performed next-generation RNA sequencing (RNA-seq) using brain tissue from 23 months old vehicle and CP2-treated non-transgenic (NTG) and Young (3 months old) WT mice. By comparing transcriptomic data of vehicle and CP2-treated NTG_Old mice, we found processes affected by CP2 such as genes involved in regulation of circadian rhythm, immune system, oxidant detoxifications. In comparison to Young mice, CP2 treatment in NTG_Old mice downregulated the expression of gene sets involved in regulation of oxidative stress and lipid metabolism, which were both upregulated with old age in vehicle-treated NTG_Old mice.
