Contrasting patterns of expression divergence in early and late stages of population divergence in mice

Project description:We compared gene expression differences in the polytypic species complex Mus musculus (Mus musculus musculus, Mus musculus domesticus, Mus musculus castaneus and Mus musculus ssp) with that of Mus spretus via oligonucleotide microarrays representing more than 20,000 genes. Analysis of the results by two way ANOVA statistics suggests that the most genes with significant differences in expression levels among the subspecies are found in liver and kidney and the least in testis. This picture is different when one compares with Mus spretus, where the largest number of differences is found in testis. The design we employed is a reference design. All tissues were hybridized against a pool of that same tissue from 9 C57BL6 mice. All mice were roughly 12 weeks of age. To control for biological variation, we have used several individual males from each sub-species. RNA was isolated from three different organs, namely brain, liver/kidney and testis.

Selective Involvement of PRC2 in Mouse Hypothalamus Cell Specification

Project description:Neuronal sub-type diversity in the hypothalamus is staggeringly immense. To investigate the role of epigenetic regulation in hypothalamic development in the generation of this cellular diversity; a floxed Eed allele was crossed to the early, pan-CNS Cre deleter Sox1-Cre, resulting in loss of H3K27me3 in the entire Mus musculus CNS by E11.5. The E18.5 embryonic hypothalamus was dissected and analysed by single cell RNA sequencing, on the Illimina/BioRad platform. Cell specification was surprisingly unaffected, with the majority of cell types - except Hcrt and dopamine neurons - sucessfully generated. This indicates that the role of the PRC2 complex - past E11.5 - is surprisingly limited in Mus musculus hypothalamic development.

RNA-seq of mouse myeloma cell line SP 2/0 overexpressed with Gm40600 against vector control

Project description:We found that some novel genes such as Gm40600 mRNA significantly changed in plasma cells. Because of plasmablast-like, mus musculus myeloma SP 2/0 cell line was selected to test the effect of Gm40600 overexpression on plasmablast/plasma cells. Gm40600 cDNA was cloned into lentiviral vector LV201 to generate Gm40600 protein. Gm40600-expressing LV201 was then transfected into SP 2/0 cells, and stable transfectants were identified by drug selection. To identify the effect of Gm40600 overexpression on gene expression, we determined mRNA profiles in Gm40600-overexpressed and vector-transduced SP 2/0 cells by RNA-sequencing. RNA-seq was done with an Illumina HiSeq 2500 instrument at GENEWIZ, Suzhou, China.
