Transcription profiling by array of chicken anterior and posterior wing bud thirds from stage HH24 normal and talpid3 mutant embryos

Project description:Relative levels of RNA transcripts were compared between anterior and posterior wing bud thirds from stage HH24 normal and talpid3 mutant chicken embryos using chicken Affymetrix chips. Data collected with Affymetrix scanner was normalized using the Plier algorithm within the expression console package from Affymetrix and log2 transformed. 5 replicates of anterior third normal wing buds, 4 replicates of posterior third of normal wing buds and 4 replicates each of anterior and posterior thirds of talpid3 wing buds at stage HH24 were examined.

Project description:This experiment was designed to investigate how inhibition of Shh signalling result in loss of flight feathers, and to identify genes involved in flight feather development. We carried out cyclopamine treatment of wing buds at HH19 which gives rise to 2 digit wings with absence of flight feathers. The posterior forewing regions of control and cyclopamine treated wings were analysed at E10.5, the developmental stage at which flight feathers first become apparent. Control leg tissue was also included in the analysis to enrich data for genes involved in general feather development. To enrich further for genes with roles in flight feather development, we also included samples from the feathered legs of Pekin bantam chickens. 3 biological replicates of pooled wing/leg sections were analysed and 12 samples were sequenced.

Comparative analysis of Gallus gallus cecal epithelia following Eimeria tenella infection

Project description:Relative expression levels of mRNAs in chicken cecal epithelia experimentally infected with Eimeria tenella were measured at 4.5 days post-infection. Two weeks old chickens were uninfected (negative control) or were orally inoculated with sporulated oocysts of Eimeria tenella. Cecal epithelia samples were collected from >12 birds in infected or uninfected group at 4.5 d following infections, in which samples from 4 birds were pooled together to form a total 3 biological replicates in each group. Parasite merozoites were also collected from four infected chickens at 5 d after infections. Uninfected control samples, merozoites and infection group samples were selected for RNA extraction and hybridization on Affymetrix microarrays. We used Affymetrix GeneChip chicken genome arrays to detail the chicken cecal epithelia gene expression in the control and E. tenella-infected birds.

Project description:The expression of genes were analysed in 7th day of embryonic stage between Aseel, an indigenous slow-growing chicken, and control broiler, a fast-growing broiler chicken line. The whole embryo was collected in TRIZOL and total RNA was isolated. The expression profile of gene was determined in 64k Agilent chicken microarray chip. The Cy3 dye was used for detection. The fold change of expression was analysed in Aseel as compared to broiler chicken line.

Anole ocular patterns of gene expression

Project description:For the purpose of facilitating gene annotation, a transcriptome profile of the developing anole eye was used to identify coding and non-coding regions in the Anolis genome. RNA datasets were generated by collecting samples from three subregions of the anole posterior globe (central, temporal, and nasal) which was comprised of nerual retina, retinal pigmented epithelium, choriod, and scleral tissues. A total of five sample replicates were made. Each replicate incorporated pooled tissues collected from three stage 16.5 developing anole embryos.
