Project description:Seasonal influenza causes significant morbidity and mortality worldwide, with 70-90% of these deaths occurring in individuals aged 65 or older. To explore the mechanisms behind this age-related vulnerability, we investigated immune responses and inflammation in the lungs of young and old mice during influenza A virus (IAV) infection. Young (12-week) and old (70-week) C57BL/6J mice were intranasally infected with IAV PR8, and immune responses were assessed at 7 days post-infection (dpi) using single-cell RNA sequencing (scRNA-seq).

Pathogenic role of alarmin IL-33 in a mouse model of SARS-CoV-2 infection

Project description:Male WT C57BL/6j mice (stock #000664, age 8-10 wks) and IL-33 knockout (KO) mice (B6 background) were anesthetized via isoflurane, followed by the intranasal infection with mouse adapted SARS-CoV-2 CMA3p20 (5×10^5 PFU). Mock mice were administrated with identical volumes of cell culture medium. Lungs were perfused with cold PBS and were harvested at days 2 and 4 post-infection. Lung RNA was isolated by Qiagen RNeasy Kits, followed by the Poly (A) RNA sequencing. We then performed differential expression analysis, meta-analysis, and gene set enrichment anaylsis using data obtained by RNA-seq of mock, IL-33 KO, and WT SARS-CoV-2-infected mice.

Gene expression changes in liver upon LCMV Cl13 infection

Project description:2-3 month old female C57Bl/6J mice were intravenously infected with 2x10^6 focus forming units of lymphocytic choriomeningitis virus strain Clone-13. Whole liver tissue was harvested in triplicates from uninfected control mice, uninfected control 123d old mice, and from infected mice at 2, 8, 30, 60, and 123 days post infection. Samples were subsequently analyzed on a HiSeq 2000 instrument using a 50 bp SE RNAseq protocol.

Twenty-week-old C57BL/6 mice were infected by intranasal instillation of 10^2, 10^3, 10^4 or 10^5 PFU of SARS CoV MA15 in 50 µl of PBS or mock-infected with PBS alone. At days 1, 2, 4 and 7 days post-infection, lungs were harvested and total RNA was isolated and subjected to microarray analysis.
