Transcriptomic analysis of human foetal hearts with or without Down Syndrome

Project description:We performed bulk RNAseq on whole hearts from foetuses with Down syndrome and age-matched, sex-matched euploid controls. Analysis showed the expected increased expression of the genes on Hsa21 that are present in 3 copies. Gene set enrichment analysis identified pathways that are altered in DS hearts.

Serum proteomics in a cohort of COVID patients and healthy controls

Project description:This project investigates the serum proteome patterms in recovered COVID patients compared to that in healthy controls. Specifically, serum saamples from 29 recovered patients and 29 age, sex, and race matched healthy controls were collected and subject to DIA-MS proteomics. A fraction of the serum from each sample was pooled for all 58 samples and run as a quality control.

Project description:In order to assess potential alterations in microRNA expression in MS patients, we first applied microRNA array to screen the expression profile of 7 controls and 6 patients in the peripheral blood mononuclear cells (PBMC) PBMC when compared to age- and sex-matched controls Peripheral blood mononuclear cells (PBMC) were prepared by density gradient centrifugation using Nycoprep 1.077 medium (Accurate Chemical & Scientific Corp, Westbury, NY, USA). Total RNA including microRNAs were extracted using miRNeasy mini kit (Qiagen, Germantown, MD, USA) and subjected to microRNA high-throughput array with Affymetrix platform. MicroRNA array in Affymetrix 3.0 platform. Quantitative real-time PCR (QPCR) was applied for the validation and amplification of the test samples. Relative fold expression values were calculated based on the expression of Snord96A gene as an endogenous control. Microarray data are available in the ArrayExpress database (HUwww.ebi.ac.uk/arrayexpressUH) under accession number E-MEXP-4673.
