Transcriptomic analysis of differentially expressed genes in the Winnie mouse model of chronic colitis

Project description:The Winnie mouse, carrying a missense mutation in the Muc2 gene, is a model for chronic colitis closely resembling the pathological changes of human IBD. Herein, transcriptomic analyses of the differentially expressed genes in the distal colon of Winnie mice with mild colitis and Winnie-Prolapse mice with severe colitis are compared to control C57BL/6 mice. Gene ontology analysis and KEGG pathway analysis demonstrated the upregulation of genes in immune and inflammation-related pathways, metabolic pathways, cancer-related pathways, and neurological processes. Further research into these pathways and individual genes may lead to the identification of new targets for the treatment of IBD. The overexpression of oncogenes, in particular, may serve as an indicator of inflammation progressing to cancer.

The gut microbiota affects the mucus barrier

Project description:Two C57BL/6 mice colonies maintained in two rooms in the same specific pathogen free (SPF) facility were found to have different gut microbiota and a mucus phenotype specific for each colony. The thickness and growth of the colon mucus was similar in the two colonies, but one colony had mucus not penetrable to bacteria or bacterial-sized beads, similar to what occurs in free-living wild mice. On the other hand, the other colony had an inner mucus layer that was penetrable to bacteria and beads. These different properties of the mucus in the two rooms were dependent on the microbiota, as the phenotypes were transmissible by transfer of ceacal microbiota to germ-free mice. Mice with an impenetrable mucus layer had increased amounts of Erysipelotrichi, while mice with a penetrable mucus layer had higher levels of Proteobacteria and TM7 bacteria in the distal colon mucus. Thus bacteria affect mucus barrier properties in ways that can have implications for health and disease.

Global gene expression profile of colon from wild-type and AE2-knockout mice.

Project description:The objective of this study was to identify changes in gene expression levels between wild-type and AE2-knockout colon. 7 wild-type and 7 AE2-null colon RNA samples were compared to identify genes that are differentially expressed in the colon of AE2 knockout mice. Total RNA was collected from both male and female wild-type and AE2-null colons (including both proximal and distal colon) when the mice were 18 days old. All pairs were gender- and aged-matched, and all mice were on a mixed 129 Black Swiss background. To facilitate statistical analysis and reduce any affects of Cy3 and Cy5 labeling, comparisons of two wild-type and two AE2-null samples were repeated using a dye flip.

Genes induced by IL-9 in the colon of transgenic mice

Project description:The aim of this work was to identify genes induced by IL-9 in the colon of IL-9-tarnsgenic mice (Tg5). Therefore, we performed a comprehensive study of the genes expressed in the colon of IL-9 transgenic and wild type FVB mice, taking advantage of the affymetrix microarray technology. Keywords: cytokine-induced response RNA was extracted from a piece of the distal colon from IL-9 transgenic or wild type TVB mice. RNA samples from 4 different mice were pooled for each group before hybridization on Affymetrix microarrays.
