Project description:The aneurysm clip impact-compression model of spinal cord injury (SCI) in animals mimics the primary mechanism of SCI in human, i.e. acute impact and persisting compression; and its histo-pathological and behavioural outcomes are extensively similar to the human SCI. In order to understand the distinct molecular events underlying this injury model, an analysis of global gene expression of the acute, subacute and chronic stages of a moderate to severe injury to the rat spinal cord was conducted using a microarray gene chip approach. Rat thoracic spinal cord (T7) was injured using aneurysm clip impact-compression injury model and the epicenter area of injured spinal cord was isolated for RNA extraction and processing and hybridization on Affymetrix GeneChip arrays.

Project description:Summary: Spinal cord injury (SCI) is a damage to the spinal cord induced by trauma or disease resulting in a loss of mobility or feeling. SCI is characterized by a primary mechanical injury followed by a secondary injury in which several molecular events are altered in the spinal cord often resulting in loss of neuronal function. Hypothesis: Spinal cord injury (SCI) induces a cascade of molecular events including the activation of genes associated with transcription factors, inflammation, oxidative stress, ionic imbalance, apoptosis and neuroregeneration which suggests the existance of endogenous reparative attempts. However, not all mechanisms following SCI are well known. Specific Aim: The goal of this project is to analyze the molecular events following spinal cord injury 1 cm above, below, and at the site of injury (T9), aiming at finding potential new targets to improve recovery and therapy.

Project description:Spinal cord injury (SCI) leads to severe neurological and functional impairments, yet reliable biomarkers for assessing injury severity and predicting recovery remain limited. Cerebrospinal fluid (CSF) is in direct contact with the central nervous system and provides a valuable source for detecting molecular changes after SCI. Although exosomal microRNAs and proteins are increasingly recognized as mediators of intercellular communication, the role of human CSF exosomes in SCI has not been systematically investigated. To identify exosome-based biomarkers and potential therapeutic targets, we analyzed CSF and serum exosomes from patients with acute SCI using RNA sequencing and proteomic profiling. Weighted Gene Co-expression Network Analysis (WGCNA) identified six gene modules significantly associated with injury severity and neurological recovery at three months. Proteomic analysis revealed a five-protein panel that distinguished complete from incomplete SCI and a four-protein panel that predicted neurological improvement. Additionally, fifteen CSF-specific and nine serum-specific exosomal miRNAs were identified independent of injury severity. Among ten tested miRNAs associated with neurological recovery, seven regulated astrocyte proliferation, and six promoted neurite extension and synapse formation. Overall, this study provides a comprehensive characterization of CSF exosomal miRNAs and proteins in human SCI and identifies molecular signatures associated with injury severity and recovery.

Project description:Spinal cord injury triggers extensive molecular remodeling in neural and glial compartments. To resolve these processes at cell-type-specific resolution, we performed quantitative proteomic profiling of laser-microdissected motor neurons (MNs), dorsal root ganglia (DRG), and ependymal cells (EC) from control Wistar rats and 2 or 6 weeks after complete spinal cord transection at Th10. Retrogradely traced MNs innervating the soleus (SOL; 172 ± 39 cells) and tibialis anterior (TA; 262 ± 74 cells) muscles, ECs from the central canal (L3–L6) were microdissected from spinal cord slices with Leica LMD7000 Laser Microdissection System. Bilateral DRG (L3–L5) were also isolated. Samples from five animals per group were analyzed using TMT-based LC–MS/MS, yielding 1 221 proteins in SOL MNs, 1 186 in TA MNs, 1,520 in ECs, and 5 087, 3 740, and 3 086 in DRG L3, L4, and L5, respectively. The dataset supports cell-type-specific characterization of molecular responses after spinal injury.

Analysis of the areas directly (spinal cord) and indirectly (raphe and sensorimotor cortex) affected by injury will help understanding mechanisms of SCI. Hypothesis: Areas of the brain primarily affected by spinal cord injury are the Raphe and the Sensorimotor cortex thus gene expression profiling these two areas might contribute understanding the mechanisms of spinal cord injury. Specific Aim: The project aims at finding significantly altered genes in the Raphe and Sensorimotor cortex following an induced moderate spinal cord injury in T9.

Project description:Although axon regeneration can now be induced experimentally across anatomically complete spinal cord injury (SCI), restoring meaningful function after such injuries has been elusive. This failure contrasts with the spontaneous, naturally occuring repair that restores walking after severe but incomplete SCI. Here, we applied projection-specific and comparative single-nucleus RNA sequencing to uncover the transcriptional phenotype and connectome of neuronal subpopulations involved in natural spinal cord repair. We identified a molecularly defined population of excitatory projection neurons in the thoracic spinal cord that extend axons to the lumbar spinal cord where walking execution centers reside. We show that regrowing axons from these specific neurons across anatomically complete SCI and guiding them to reconnect with their appropriate target region in the lumbar spinal cord restores walking in mice. These results demonstrate that mechanism-based repair strategies that recapitulate the natural topology of molecularly defined neuronal subpopulations can restore neurological functions. Expanding this principle to different classes of neurons across the central nervous system may unlock the framework to achieve complete repair of the injured spinal cord.

Project description:The goal of this study is to elucidate the influence of hemisection injury at thoracic spinal cord (T9) on the transcriptome of the lower lumbar spinal cord at acute phase. mRNA profiles of spinal cord at 4 days-post injury and before injury were generated. 72 Differentially Expressed Genes (DEGs) were observed. Our study represents the detailed analysis of transcriptomes of spinal cord distal to the hemisected lesion at acute phase, with biologic replicates, generated by RNA-seq technology.

Project description:After spinal cord injury, fibrotic scars will form, which affect tissue regeneration and functional recovery in the body. However, the differences in fibrotic scars produced by different segments of the spinal cord are still unclear. Here, we demonstrate that after the same spinal cord hemisection injury, the thoracic spinal cord produces the most scars, while the lumbar and cervical spinal cords have fewer scars, and there are also significant differences in functional recovery after injury in the three segments. By comparing the differences in fibrosis scars produced by cervical spinal cord, thoracic spinal cord, and lumbar spinal cord, as well as the different abilities of fibroblasts to promote fibrosis in vitro, the potential mechanism characteristics of injury repair in different segments of the spinal cord were revealed. This article explores the impact of spinal cord segment differences on fibrosis scar formation and their relationship with behavioral functional recovery, revealing the regional heterogeneity of spinal cord tissue fibrosis scar formation and deepening our understanding of the complexity of CNS scar formation. This provides new ideas for future targeted treatment to improve functional recovery after specific spinal cord injuries.

Project description:Recent studies in brain and spinal cord have revealed the heterogenous nature of astrocytes; however, how diverse constituents of astrocyte lineage cells are regulated in adult spinal cord after injury and contribute to regeneration remains elusive. We performed single-cell RNA-sequencing (scRNA-seq) of astrocyte lineage cells from sub-chronic spinal cord injury (SCI) models, identified and compared with the subpopulations in the acute stage data. We found the subpopulations with distinct functional enrichment and their identities defined by subpopulation-specific transcription factors and regulons. Our analyses revealed the molecular signature, location and morphologies of potential residential neural progenitors or neural stem cells in the adult spinal cord before and after injury, and the intermediate cells enriched in neuronal markers that could potentially transition into other subpopulations. The investigation of stage-specific cell-cell communications among astrocyte lineage cells and with other cell types in the tissue generated valuable insight into signaling pathway networks in SCI. This study has significantly expanded the knowledge of the heterogeneity and cell state transition of glial progenitors in adult spinal cord before and after injury.
