Expression data from Arabidopsis wild-type, sicy-192, and sicy-192 gun1-101 plants

Project description:Plastid gene expression (PGE) acts as a signal that regulates the expression of photosynthesis-associated nuclear genes (PhANGs) via GENOMES UNCOUPLED1 (GUN1)-dependent retrograde signaling. We recently isolated Arabidopsis sugar-inducible cotyledon yellow-192 (sicy-192), a gain-of-function mutant of plastidic invertase (INV-E), and showed that following the treatment of this mutant with sucrose, the expression of PhANGs decreased while that of nitrate reductase 1 (NIA1) increased. Plastid-encoded RNA polymerase (PEP)-dependent PGE was markedly suppressed in the sicy-192 mutant by the sucrose treatment. A double mutant of sicy-192 and gun1-101, a null mutant of GUN1, revealed that metabolic perturbation in the sucrose-treated sicy-192 mutant was, at least in part, dependent on GUN1. To confirm whether there is a relationship between plastid sugar metabolism and nuclear gene expression, we performed a microarray analysis using Suc-treated 3 genotypes consisting of wild-type, sicy-192, and sicy-192 gun1-101 plants.

Transcription profiling to investigate the photoperiodic control of starch mobilisation in Arabidopsis thaliana

Project description:In order to identify genes specifically involved in the photoperiodic control of the mobilisation programme for starch reserve in Arabidopsis thaliana transcription profiling was performed on the following genotypes Columbia wild type (Col-0), CO overexpressor (35S::CO), CO muntant (co-10), GBSSI mutant (gbs-1), aps1 mutant (aps1) and Columbia with sucrose. The different Arabidopsis thaliana genotype seedlings were cultivated in long day conditions (16 h day/ 8 h dark) at 22 C in controlled environment cabinets for two weeks. Samples were collected at ZT 4, immediately frozen in liquid nitrogen and processed.

Gene expression in response to genetic and chemical perturbations of chromatin structure

Project description:Microarray expression profiling was used to identify genes expressed misexpressed in wild-type Arabidopsis seedlings treated with 5-aza-2’ deoxyctidine (5AC) or trichostatin A (TSA), and in decrease in dna methylation1 (ddm1) mutant seedlings. 10 day-old Arabidopsis seedlings (ecotype Ws) were grown in 0.5X MS media (containing 1% sucrose) at 21 C in continuous white light. Media (including 5AC and TSA) were refreshed after 5 days of growth. RNA was extracted using the Qiagen Plant RNeasy kit. Three biological replicates for each treatment and genotype were used.

Identification of early gibberellin (GA) responsive genes in Arabidopsis hypocotyls

Project description:The aim of the experiment was to identify early gibberellin (GA) responsive genes in the roots of an Arabidopsis GA deficient mutant.The GA deficient mutant used in this study is a transgenic line overexpressing the PcGA2ox1 gene. This mutant has an identical phenotype to ga1-3, but it does not require exogenous GA treatment for germination.Seeds were germinated on 1xMS + 1% (w/v) sucrose plates containing 0.7% gelrite, and grown under continous light. The plates were orientated vertically.After six days growth the plates were treated with or without 5uM GA4 for 0, 30, 60 and 180 minutes.Approximately 400 hypocotyls were harvested per timepoint using a razorblade and after excision the hypocotyls were frozen in liquid nitrogen giving a total of 7 experimental samples (1: untreated, 2: 30 mins GA4, 3: 30 mins untreated, 4: 60 mins GA4, 5: 60 mins untreated, 6: 180 mins GA4, 7: 180 mins untreated. Three biological replicates were performed giving a total of 21 root samples. Total RNA was isolated from the roots using the QIAGEN RNeasy method. Keywords: Expression profiling by array
