Project description:In this experiment, mice from 3 strains A/J, Balb C and C57Bl6 were infected with Trypanosoma Congolense, IL1180 clone, African sleeping sickness, a disease which affects cattle in sub-saharan Africa. These mouse strains were chosen because they are a model for tolerance to infection in cattle, with A/J and Balb B being highly susceptible, and C57Bl6 being somewhat more tolerant to infection. Three tissues Kidney Liver and Spleen were harvested from cohorts at various timepoints: 0 (naive), 3, 7, 9 and 17 days post infection. Each condition thus comprises: Time, Tissue, Strain. For each condition, RNA extracts from individuals were pooled (5 per pool) prior to hybridisation.

Project description:Tissues were harvested from mice from two congenic strains (Tir1CC and Tir3CC) and their two control strains (Tir1AA and Tir3AA). The congenic strains were bred from inbred lines known to be susceptible (A/J) and tolerant (C57BL/6) to Trypanosome infection, such that C57BL/6 genomic regions underlying QTL for trypanotolerance were present in an A/J genetic background. Liver and spleen tissues were harvested. For each strain RNA was prepared from various tissues of twenty mice. For each condition, RNA extracts from individuals were pooled (5 per pool) prior to hybridisation.

Project description:In this experiment, cattle from 2 breeds Boran and N'Dama were infected with Trypanosoma Congolense, IL1180 clone, African sleeping sickness, a disease which affects cattle in sub-saharan Africa. These breeds were chosen because they differ in their tolerance to infection, with Boran being highly susceptible, and N'Dama being more tolerant to infection. Three tissues Liver, Spleen and Lymph Node were harvested from individuals at various timepoints. Liver, Spleen and Lymph Node were collected post mortem at days 0 (naive), 21 and 35 post infection. In addition, further liver samples were collected by biopsy at days 0 (naive), 12, 15, 18, 26, 29, 32 days post infection. Each condition thus comprises: Time, Tissue, Breed. For each condition, RNA extracts from individuals were hybridised on individual arrays.<br>Infection experiment, tissue harvesting and RNA extraction were carried out at ILRI Nairobi Kenya.<br>Hybridizations were carried out at the Microarray facility, Roslin Institute, UK.<br>Data analysis was carried out at Liverpool University and Manchester University, UK.<br>

Project description:Kaiser2014 - Salmonella persistence after ciprofloxacin treatment
The model describes the bacterial tolerance to antibiotics. Using a mouse model for Salmonella diarrhea, the authors have found that bacterial persistence occurs in the presence of the antibiotic ciprofloxacin because Salmonella can exist in two different states. One, the fast-growing population that spreads in the host's tissues and the other, slow-growing "persister" population that hide out inside dendritic cells of the host's immune system and cannot be attacked by the antibiotics. However, this can be killed by adding agents that directly stimulate the host's immune defense.

Abstract:
In vivo, antibiotics are often much less efficient than ex vivo and relapses can occur. The reasons for poor in vivo activity are still not completely understood. We have studied the fluoroquinolone antibiotic ciprofloxacin in an animal model for complicated Salmonellosis. High-dose ciprofloxacin treatment efficiently reduced pathogen loads in feces and most organs. However, the cecum draining lymph node (cLN), the gut tissue, and the spleen retained surviving bacteria. In cLN, approximately 10%-20% of the bacteria remained viable. These phenotypically tolerant bacteria lodged mostly within CD103⁺CX₃CR1⁻CD11c⁺ dendritic cells, remained genetically susceptible to ciprofloxacin, were sufficient to reinitiate infection after the end of the therapy, and displayed an extremely slow growth rate, as shown by mathematical analysis of infections with mixed inocula and segregative plasmid experiments. The slow growth was sufficient to explain recalcitrance to antibiotics treatment. Therefore, slow-growing antibiotic-tolerant bacteria lodged within dendritic cells can explain poor in vivo antibiotic activity and relapse. Administration of LPS or CpG, known elicitors of innate immune defense, reduced the loads of tolerant bacteria. Thus, manipulating innate immunity may augment the in vivo activity of antibiotics.

Project description:Ascariasis is a neglected tropical disease that affects 800 million people worldwide. Whereas most people only experience light worm burden, some people experience heavy worm burdens even after several rounds of chemotherapy, a phenomenon known as predisposition. Such heavy infections are associated with more severe symptoms and increased chronic morbidity. In order to investigate potential mechanisms that may explain the observed predisposition, we infected mice with the porcine ascarid A. suum using an established mouse model with two different mouse strains, C57BL/6J and CBA/Ca, as a model for heavy and light infection. At day 7 post infection we investigated the liver proteome, using shotgun mass spectrometry, of both infected and control mice of each strain. We identified intrinsic differences, between the two mouse strains, in both oxidative phosphorylation proteins and proteins involved in the retinol metabolism. Additionally, we found differences between the two mouse strains in activation of the complement system, where the resistant mouse strain has higher protein abundances for lectin pathway proteins and the susceptible mouse strain has higher protein abundances for complement inhibiting proteins. The resistant mouse strain had a higher abundance of proteins involved in the activation of the complement cascade via the lectin pathway. In contrast, the susceptible strain demonstrated a higher abundance of proteins involved in arresting the complement pathway. In conclusion, we observed clear differences between the two mouse strains both intrinsically and under infection.

Project description:Ectromelia virus (ECTV) has emerged as a valuable model for investigating the host-orthopoxvirus relationship as it relates to pathogenesis and the immune response. ECTV causes mousepox in most strains of mice, including BALB/c and DBA/2, and these are therefore classified as susceptible mice. Conversely, C57BL/6 and certain 129 strains display limited pathology and a very low mortality, and are thus classified as resistant. To understand the host genetic factors of different mouse strains in response to ECTV infection, we carried out a microarray analysis using Affymetrix Gene-Chip Mouse Genome Arrays of spleen tissues from BALB/c and C57BL/6 mice at 3 and 10 days post-ECTV infection. Differential Expression of Genes (DEGs) analyses revealed distinct differences in the gene profiles of resistant and susceptible mice infected with ECTV. Gene ontology and KEGG pathway analysis showed that the DEGs of susceptible mice were involved mainly in immunity, apoptosis, spliceosomes and cancer-related pathways, while the DEGs of resistant mice were largely involved in MAPK signaling and leukocyte transendothelial migration. This suggests that the susceptible BALB/c mice have a stronger response than the resistant C57BL/6 mice to ECTV infection. The BALB/c mice showed a strong induction of interferon-induced genes (ISGs), including guanylate binding proteins (GBPs), myxovirus resistance protein (Mx) GTPases, oligoadenylate synthetase (OAS) and IFN-induced protein with tetratricopeptide repeats (IFIT) family proteins, while the C57BL/6 mice upregulated more genes related to metabolic pathways. This suggests that the susceptible BALB/c mice have a stronger response than the resistant C57BL/6 mice to ECTV infection.

Project description:Amphibian populations around the world are threatened by an emerging infectious pathogen, the chytrid fungus Batrachochytrium dendrobatidis (Bd). How can a fungal skin infection kill such a broad range of amphibian hosts? And why are certain species particularly susceptible to the impacts of Bd? Here we use a genomics approach to understand the genetic response of multiple susceptible frog species to Bd infection. We characterize the transcriptomes of two closely-related endangered frog species (Rana muscosa and Rana sierrae) and analyze whole genome expression profiles from frogs in controlled Bd-infection experiments. We integrate the Rana results with a comparable dataset from a more distantly-related susceptible species (Silurana tropicalis). We demonstrate that Bd-infected frogs show massive disruption of skin function and show no evidence of a robust immune response. The genetic response to infection is shared across the focal susceptible species, suggesting a common effect of Bd on susceptible frogs. A total of five (12-plex) chips were analyzed from 60 samples comprising 2 conditions (control and infected), 3 tissues (skin, liver and spleen) and 2 timepoints (early and late). Three biological replicates were used for each condition and tissue at each time point. Twentyfour arrays were analyzed for skin samples, 24 for liver, and 12 for spleen. The same dye, Cy5, was used for all samples.
