ABSTRACT: DIDO3 colocalization with the epigenetic markers H3K4me3, H3K36me3, H3K9me3, and H3K27me3. DIDO3 a.a. 1-528 with the PHD mutation C285A/H290A (HA-DIDO3-PHD) or with the wild-type PHD domain (HA-DIDO3-NT) were employed.

The impact of bacterial binding dressings following colorectal surgery on surgical wound complications in an adult population.

Project description:Interventions: Trial participants will be allocated to either the intervention or control group. There is currently only one DACC dressing (Leukomed Sorbact) available in the wound care dressing. The intervention group will receive the DACC containing dressings following surgery (Leukokmed Sorbact). The DACC dressing is a cover dressing applied over the incision site The control group will receive a dressing that does not contain DACC and has the same dressing properties without the interventional dressing characteristics. Participants allocated to the control group will receive a non DACC dressing. The wear time for both groups will be 14 days, unless otherwise clinical indicated for a dressing change and assessment by the lead PI. Dressing application and wear time is to be conducted according to the manufacturer specifications. Dressings will be applied as per standard of post operative care. Dressings will be visually checked daily by the study lead wound care clinician. Primary outcome(s): The primary outcome will be determined by clinical assessment and diagnosis as well as patient reported outcome. Post discharge surveillance using telemedicine and a phone call at 3o days will be conducted on all trial participants. The primary outcome measurement is defined as the occurrence of surgical site infection as per Centres of Disease Control definition and coding ICD-11 T81.49. [Day 30 postoperative ];The primary outcome measurement is defined as the occurrence of surgical wound dehiscence as defined by World Union of Wound Healing Societies definition and coding ICD 11: T81.3. Post discharge surveillance will be conducted using telemedicine and a phone call at 30 days after surgery. Whole genome sequencing of core-binding factor leukemia

Project description:Pediatric de novo acute myeloid leukemia (AML) is a heterogeneous disease that can be divided into clinically distinct subtypes based on the presence of specific chromosomal abnormalities or gene alterations. One of the best characterized subtypes of AML involves leukemias with alterations of the core-binding factor (CBF)-complex, which comprises the FAB subtypes M2 and M4Eo and associates with a favorable outcome. Patients with the AML M2 subtype harbor a translocation between chromosomes 8 and 21 [t(8;21)] that yields the chimeric fusion gene RUNX1(AML1)-RUNX1T1(ETO), while patients with AML M4Eo express the chimeric fusion gene CBFβ-SMMHC(MYH11) as a result of an inversion/translocation event of chromosome 16 [inv(16)/t(16;16)]. In an effort to define the total complement of genetic changes in CBF-leukemia, we performed paired-end whole genome sequencing (WGS) on diagnostic leukemia blasts and matched germ line samples from 17 pediatric CBF-leukemia patients using the Illumina platform. Somatic alterations, including single nucleotide variations (SNVs) and structural variations (SVs), including insertions, deletions, inversions, and inter- and intra-chromosomal rearrangements, were detected using complementary analysis pipelines (Bambino, CREST and CONSERTING). Recurrent screening of identified mutations will be performed in a cohort of approximately 94 cases of CBF-leukemias.
