Live-cell analyses with unsegmented images to study cancer cell response to modified T cell therapy

ABSTRACT: RASA2KO and WT TCR T cells co-cultured with A375 melanoma cell line. The A375 cells express RFP which has the highest intensity in the nucleus. The TCR T cells are engineered to target the NY-ESO-1 antigen that is expressed by the A375 cells. Each well is imaged once every 2hrs using the automated Incucyte Zoom live-cell imaging machine. Phase and red channels are collected for each image. Spatial resolution is 4.975 μm/pixel.
