LeGO-3D: 3D imaging of lung metastases and vascularisation using light sheet fluorescence microscopy
Ontology highlight
ABSTRACT: Cancer metastasis involves a complex cascade of events, where cancer cells migrate from their site of origin to secondary sites via the lymphatic and circulatory system. Mouse models of advanced metastatic breast cancer are usually established orthotopically in the mammary fat pad (MFP) or by intravenous (IV) injection of cell lines (e.g. MDA-MB-231) or via patient-derived xenografts (PDX). These models of metastatic disease are critical for understanding the drivers of clonal diversity and drug resistance, as intra-tumoral heterogeneity is a characteristic of poor clinical outcomes. To understand the growth of metastatic tumours in tissue microenvironments, we adapted and optimised a method based on lentiviral-gene ontology optical barcoding known as LeGO for 3-dimensional imaging of whol
