Project description:To further gain insights into the role of Bdh2 in cell fate decisions of mouse ESCs, we generated Bdh2 homozygous knockout mouse advanced pluripotent embryonic stem cell (ASC) and embryonic stem cell (ESC) lines by CRISPR/Cas9 genome editing technology. Bdh2 deficiency in ASCs/ESCs exhibited no effect on expression of core pluripotent transcription factors and alkaline phosphatase activity, suggesting dispensability of Bdh2 for self-renewal and pluripotency of embryonic stem cells. Notebly, Bdh2-knockout ASCs/ESCs exhibit higher expression of the endodermal somatic genes, Gata4, Gata6, Sox17,and also colocalizated with Oct4. Importantly, expression of key DNA methylation genes Dnmt3a, Dnmt3b and Dnmt1 decreased in Bdh2-knockout ASCs/ESCs. Overall, we provide insight on understanding of earliest cell fate decicion in ASCs/ESCs and a new avenue to create and preserve pioneer lineage precursors with pluripotency.

Opposing roles for AMPK in regulating distinct mitophagy pathways

Project description:Mitophagy degrades damaged mitochondria, but we show here that it can also target functional mitochondria. This latter scenario occurs during programmed mitophagy and involves the mitophagy receptors NIX and BNIP3. While AMPK, the energy-sensing protein kinase, can influence damaged-induced mitophagy, its role in programmed mitophagy is unclear. We found that AMPK directly inhibits NIX-dependent mitophagy by triggering 14-3-3-mediated sequestration of ULK1, via ULK1 phosphorylation at two sites: Ser556 and an additional identified site, Ser694. In contrast, AMPK activation increases Parkin phosphorylation and enhances the rate of depolarisation-induced mitophagy, independently of ULK1. We show that this happens both in cultured cells and tissues in vivo, using the mito-QC mouse model. Our work unveils a mechanism whereby AMPK activation downregulates mitophagy of functional mitochondria but enhances that of dysfunctional/damaged ones.
