Time-lapse ND2 microscopy dataset of E. coli under IPTG and Plain M9 perturbations


ABSTRACT: This dataset contains long-term, multi-dimensional time-lapse microscopy acquisitions of Escherichia coli grown in microfluidic chambers under defined environmental perturbations. Cells were monitored over approximately 100 hours using phase-contrast and fluorescence microscopy to quantify promoter activity dynamics at single-cell resolution. Experimental conditions include pre-conditioning phases and medium perturbations with 1 mM IPTG or Plain M9, enabling controlled induction and comparative analysis of gene expression responses. The dataset consists of raw Nikon ND2 files acquired sequentially and designed to capture cellular growth, division, and fluorescence dynamics over extended periods. These data were used to evaluate segmentation-based single-cell fluorescence quantification and to benchmark computational analysis workflows implemented in the Partaker software platform. The dataset enables analysis of population heterogeneity, temporal expression shifts, and perturbation-induced transitions in microbial communities. All files are provided in their original format to ensure reproducibility and compatibility with downstream image analysis pipelines.
