SARS-CoV-2 infection dynamics in lungs of African green monkeys

Project description:Detailed knowledge about dynamics of SARS-CoV-2 infection in vivo is important for unraveling the viral and host response factors that contribute to COVID-19 pathogenesis. The unknown dose and exposure timing in human infections makes the needed well-controlled time course studies impossible and thus animal models of disease are essential to fill in the gaps in our understanding of disease progression. Old-World nonhuman primate species recapitulate mild COVID-19 cases, thereby serving as important models for studying disease pathogenesis. In this study, we compare African green monkeys (AGM; Chlorocebus sabaeus) inoculated with SARS-CoV-2 to AGM inoculated with a gamma-irradiated form of the virus to study the dynamics of virus replication throughout the respiratory tract and other target tissues. RNA sequencing of single cells from the lungs and mediastinal lymph nodes allowed a high-resolution, simultaneous analysis of virus replication and the host response in these tissues over time. Viral replication was mainly localized to the lower respiratory tract, especially the pneumocyte population. However, even in the absence of active replication, viral genomic RNA is was highly stable, especially in the upper respiratory tract. Macrophages play a vital and dynamic role in initiating a pro-inflammatory state in the lungs, while also interacting with infected pneumocytes. Together, our dataset provides a detailed view of changes in host and virus replication dynamics over the course of a mild COVID-19 infection and serves as a valuable resource to identify new therapeutic targets.

In order to do so we injected Nippostrongylus brasiliensis (Nb) in recipient mice as this is known to elicit a Type 2 response. Mice were subcutaneously injected with Nb larvae and single CD4+ cells were isolated from mediastinal lymph nodes, mesenteric lymph nodes, and lungs 5 days after infection.

Project description:To better understand the biological pathways by which UV inactivated SARS-CoV-induced pulmonary eosinophilia occurs, we examined global transcriptional changes in macrophages from the lungs of mouse. Female BALB/c mice were used at 21 weeks of age. Mice were subcutaneously immunized with UV inactivated SARS-CoV (UV-V) or UV-V and Toll like receptor (TLR) ligands at 6 and 1 weeks prior to mouse-adapted SARS-CoV (n=6 per group). Mice were intranasally challenged with 1E+6 TCID50 in 30M-NM-<L. MEM was challenged in six mice as control infection. Mice were sacrificed and collected lungs at 1 days after challenge, then CD11b positive cells were isolated from the lungs of these mice. These cells were used for the analysis of microarray.

Soluble lymph node proteome during anthrax infection in mice

Project description:This study aimed to characterize a soluble proteome of popliteal lymph nodes during lymphadenitis induced by intradermal injection of Bacillus anthracis Sterne spores in mice using tandem LC-MS/MS. More than 380 proteins were detected in the normal intra-nodal lymph, while the infectious process resulted in the profound changes in the protein abundances and appearance of 297 unique proteins. These proteins belong to an array of processes reflecting response to wounding, inflammation and perturbations of hemostasis, innate immune response, coagulation and fibrinolysis, regulation of body fluid levels and vascular disturbance among others. The results demonstrate that the proteome of intra-nodal lymph serves as a sensitive sentinel of the processes occurring within the lymph nodes during infection. The acute innate response of the lymph nodes to anthrax is accompanied by cellular damage and inflammation with a large number of up- and down-regulated proteins many of which are distinct from those detected in serum.

Caspase-11 promotes inflammation and thrombosis in SARS-CoV-2 infection

Project description:SARS-CoV-2 infections are a worldwide health concern, and new treatment strategies are needed for decreasing virus-induced inflammatory tissue damage. Targeting inflammatory innate immunity pathways holds therapeutic promise, but effective molecular targets remain elusive. Here, we show that the innate immunity proteins, human caspase-4 (CASP4), and its mouse homologue, caspase-11 (CASP11), are upregulated in SARS-CoV-2 infections, and that CASP4 expression correlates with severity of SARS-CoV-2 infection in humans. SARS-CoV-2-infected Casp11-/- mice experienced less severe infections in terms of weight loss and lung damage than WT mice. Notably, these phenotypes were not recapitulated in mice lacking the CASP11 downstream effector gasdermin D (Gsdmd-/-), though viral titers were similar in all groups. Global transcriptomics of infected WT and Casp11-/- lungs identified decreased inflammation and neutrophil gene signatures. We confirmed that protein levels of inflammatory mediators IL-1β and CXCL1, and neutrophil infiltration, were decreased in Casp11-/- lungs. Additionally, Casp11-/- lungs expressed less von Willebrand factor, a marker for endothelial dysfunction and more Kruppel-Like Factor 2 (KLF2), a transcription factor with anti-thrombotic functions. Thus, CASP11 is established as an upstream regulator of blood coagulopathy in SARS-CoV-2 infection. Overall, our results demonstrate that CASP11, promotes detrimental SARS-CoV-2-associated inflammation and coagulopathy, identifying CASP11 as a promising drug target for treatment and prevention of tissue damage in COVID-19.
