Liu2025 - Models of Oncolytic Virus Therapy in Zebrafish


ABSTRACT: (First submission) This submission encompasses a suite of three interconnected models that aim to describe oncolytic virus therapy in zebrafish. The baseline model establishes the fundamental dynamics by modeling uninfected and infected tumor cells and extracellular virus particles with ordinary differential equations, providing an initial framework for tumor–virus interactions. However, to capture the experimentally observed delay in tumor shrinkage, the age‐of‐infection model was developed; this model introduces multiple infected cell stages to represent the gradual increase in viral load prior to cell lysis. Finally, the individual‐based extension refines the model by incorporating variability in tumor cell homing among zebrafish, thereby addressing inter-individual differences in treatment responses. Together, these models offer a comprehensive and layered approach to understanding the complex dynamics of oncolytic virus therapy, highlighting both the importance of a delayed infection mechanism and the heterogeneity inherent in biological systems.
