Transcription profiling of rat Ren2 left ventricles to generate predictive heart failure expression profile

Project description:The intercalated disc of cardiac myocytes is emerging as a crucial structure in the heart. Loss of intercalated disc proteins like N-cadherin causes lethal cardiac abnormalities, mutations in intercalated disc proteins cause human cardiomyopathy. A comprehensive screen for novel mechanisms in failing hearts demonstrated that expression of the lysosomal integral membrane protein-2 (LIMP-2) is increased in cardiac hypertrophy and heart failure in both rat and human myocardium. Complete loss of LIMP-2 in genetically engineered mice did not affect cardiac development; however these LIMP-2 null mice failed to mount a hypertrophic response to increased blood pressure but developed cardiomyopathy. Disturbed cadherin localization in these hearts suggested that LIMP-2 has important functions outside lysosomes. Indeed, we also find LIMP-2 in the intercalated disc, where it associates with cadherin. RNAi-mediated knockdown of LIMP-2 decreases the binding of phosphorylated b-catenin to cadherin, while overexpression of LIMP-2 has the opposite effect. Taken together, our data show that lysosomal integrated membrane protein-2 is crucial to mount the adaptive hypertrophic response to cardiac loading. We demonstrate a novel role for LIMP-2 as an important mediator of the intercalated disc. Transcription profiling of rat non-alcoholic Steatohepatitis following feeding of high polyunsaturated fat diets

Project description:Most commonly used models of non-alcoholic steatohepatitis (NASH) are diets based on specific gene knockouts or represent extreme manipulations of diet. We have examined the effects of modest increased caloric intake and high dietary unsaturated fat content on the development of NASH in male rats using a model in which overfeeding is accomplished via intragastric infusion of liquid diets as a part of total enteral nutrition. Male Sprague dawley rats were fed diets 5% corn oil containing diets at 187 Kcal/kg3/4/d or fed 70% corn oil containing diets at 220 Kcal/kg3/4/d for a period of 3 weeks. Hepatic gene expression were assessed at the end of the study. Our results indicate that overfeeding of high unsaturated fat diets leads to pathological, endocrine and metabolic changes characteristic of NASH patients and is associated with increased oxidative stress and TNF-a. Experiment Overall Design: Two groups of male sprague dawley rats were fed liquid diets via total enteral nutrition. Experiment Overall Design: Group 1, Control, Rats were fed diets containing 5% Corn oil at 187 Kcal/kg3/4/d for 3 weeks. Experiment Overall Design: Group 2, NASH, Rats were fed diets containing 70% corn oil at 220 Kcal/kg3/4/d for 3 weeks.

Then for the split-root experiments, the root system was washed in water to discard the sand and separated in two parts dipped in 2 containers (300 mL each) containing either the same medium (MS/10 supplied with 500 ?M Pi (cP500) or without Pi (cP0) or different media (500 ?M Pi and no Pi, respectively spP500 and spP0) for two days. Three plants were pooled for each condition and the experiment was triplicated.

Transcription profiling of rat perichondral and reserve growth plate zones

Project description:In the growth plate, the reserve and perichondral zones have been hypothesized to have similar functions, but their exact functions are poorly understood. Our hypothesis was that significant differential gene expression exists between perichondral and reserve chondrocytes that may differentiate the respective functions of these two zones. Normal Sprague-Dawley rat growth plate chondrocytes from the perichondral zone (PC), reserve zone (RZ), proliferative zone (PZ), and hypertrophic zone (HZ) were isolated by laser microdissection and then subjected to microarray analysis. In order to most comprehensively capture the unique features of the two zones, we analyzed both the most highly expressed genes and those that were most significantly different from the proliferative zone (PZ) as a single comparator. Experiment Overall Design: 2 microarray replicates were obtained for each zone, using pooled quantities of RNA from 3 rats each at 42 and 46 days of age.

Transcription profiling of rat renal carcinogenesis model - oxidative stress-induced

Project description:An iron chelate, ferric nitrilotriacetate (Fe-NTA), induces oxidative renal tubular damage that subsequently leads to renal cell carcinoma in rodents. Here we used gene expression microarrays to find target oncogenes in this model. Network analysis of the gene expression microarray data revealed the involvement of beta-catenin pathway in the induced cancers. Experiment Overall Design: Carcinogenesis protocol was performed using specific pathogen-free male Wistar rats or male F1 hybrid rats between Fischer344 and Brown-Norway strains. A total of 10 microarrays (Rat Genome 230 2.0; 31,999 genes; Affymetrix Inc., Santa Clara, CA) were used for the screening purpose; two for each group of untreated control, Fe-NTA treatment for 1 week, Fe-NTA treatment for 3 weeks, Fe-NTA-induced RCCs with neither peritoneal invasion nor metastasis and Fe-NTA-induced RCCs with pulmonarymetastasis.

For each treatment and vehicle group there are 3-5 individual samples, as well as 1 pooled sample. Experiment Overall Design: 10ug of total RNA were hybridized to Affymetrix RG_U34A arrays according to the manufacturer's protocol. Signal intensities were generated using MAS5, with global scaling set to 1500.
