MicroRNA profiling by array of Sus scrofa endothelial cells isolated from the athero-protective descending thoracic aorta and the athero-susceptible aortic arch

ABSTRACT: This study investigates the regional expression profiles of microRNAs (miRNAs) in endothelial cells isolated from the athero-protective descending thoracic aorta (DT) and from the athero-susceptible aortic arch (AA) in a swine model. Seven 2-channel assays comparing DT samples to AA samples were performed using the Invitrogen NCode Multi-Species miRNA Microarray.

Project description:This study investigates the role of endothelial cell (EC) gene expression in the focal origin of atherosclerosis. The EC transcriptome was profiled in multiple arterial regions of normal swine. Specifically, small amounts of EC RNA were isolated from 7 athero-susceptible and 6 athero-protected regions. The number of replicates for each site varied between 4 and 8. A total of 98 samples from 76 animals were used. For each sample, linearly amplified RNA (labeled with Cy5) was co-hybridized with pooled arterial reference RNA (labeled with Cy3) onto a custom-printed porcine oligo microarray (70-mers).

Transcription profiling of human aortic endothelial cells with miR-10a knock down

Project description:This study investigates the miR-10a-mediated transcriptome in human aortic endothelial cells (HAECs). The EC transcriptome was profiled in miR-10a knockdown HAECs. Specifically, small amounts of EC RNA were isolated from control cells and miR-10 knockdown cells expressing miRNA inhibitor control (Dharmacon) and miR-10a inhibitors (Dharmacon), respectively . Eight 2-channel assays comparing knockdown to control were performed using the Agilent Whole Human Genome Microarray 4x44K [G4112F].

Project description:Atherosclerosis preferentially develops in arterial regions where hemodynamic disturbed flow and oxidative stress are present. Epigenomic regulation, especially DNA methylation, plays an essential role in regulating gene expression in response to environmental factors. We investigated the DNA methylation of endothelial cells isolated from distinctly different hemodynamic and oxidative stress environments in normal adult domestic swine: an athero-susceptible site located at the inner curvature of the aortic arch (AA) and an athero-protected region in the descending thoracic aorta (DT). Genome-wide DNA methylation landscapes as well as differential methylation regions (DMRs) were generated by methylated DNA immunoprecipitation sequencing (MeDIP-seq).

Project description:Atherosclerosis preferentially develops in susceptible regions of the arterial system that are defined by the regional vascular hemodynamics. Previous work in adult castrate male pigs revealed the coexistence of pro- and anti-atherosclerotic profiles in endothelial cell gene expression in disturbed flow regions of arteries in the absence of risk factors for atherogenesis. This study investigates the impact of gender, high fat/high cholesterol diet and regional hemodynamics on arterial endothelial cell gene expression profiles in sexually mature intact pigs.
