RNA sequencing of the intra erythrocytic lifecycle of Plasmodium chabaudi

ABSTRACT: The aim of this study is to characterise gene expression in the rodent malaria intra-erthrocytic development cycle. This will inform two further studies looking at host-parasite interactions in rodent malaria and in comparison to the same stage of human malaria will provide insight into variation in lifecycles between human and rodent malaria. Project description:We have shown previously (Reid & Berriman, NAR, 2012) that by simultaneously examining host and parasite gene expression over the course of infection we can determine pairs of genes involved in host-parasite interaction. Here we are producing a high quality dataset which will specifically allow us to exploit this finding to identify genes involved in malaria host-parasite interaction. This has been done in collaboration with Jean Langhorne at NIMR. This data is part of a pre-publication release. Strongyloides ratti molecular basis of parasitism

Project description:The genus Strongyloides spp. include important human parasites. There is also a well studied rodent model, S. ratti. Uniquely among parasitic nematodes, the Strongyloides life-cycle includes both a parasitic female stage and a genetically identical free-living female stage. Differences between these two female forms must be epigenetic, presumably controlled by altered transcription and translation. This is a project to compare the proteome and transcriptome of the parasitic and free-living females of S. ratti. From this we will define the genes and gene products of the parasitic female stage. This approach exploits the currently advanced S. ratti genome sequencing project. This work will give an understanding of the molecular basis of nematode parasitism, and so define new potential drug targets. This data is part of a pre-publication release. For information on the proper use of pre-publication data shared by the Wellcome Trust Sanger Institute (including details of any publication moratoria), please see http://www.sanger.ac.uk/datasharing/ Schistosomes are blood dwelling digenean trematodes that mature as adults in the intestinal or urinary veins, predominantly of mammals. Adult female Schistosomes produce eggs of which approximately 40% fail to pass into faeces or urine (species-dependent) but are dispersed by the blood stream into different organs where they provoke severe inflammation. This parasitic infection is known as schistosomiasis and considered by the WHO as the second most socioeconomically devastating parasitic disease, next only to malaria, with hundreds of millions infected worldwide. As the eggs represent the causative pathogenic agents, the understanding of egg forming processes, and therefore of the schistosomal reproductive biology in general, is of fundamental interest. Additionally the differences between immature (pre-mating) and mature (post-mating) adult worms are of significant interest; adults only reach maturity upon mating. Trasncriptomic sequencing of gonad-specific cellular material will help to unravel signal transduction cascades involved in e.g. gametogenesis and/or vitellogenesis. This project aims to characterize the transcriptome profiles of immature and mature adult ovaries and adult testes.

Project description:Total RNA was extracted from adult male and female zebrafish. Illumina cDNA libraries were prepared from polyA+ RNA and sequenced using Illumina HiSeq paired-end sequencing.
