Transcriptomic study of haemopoietic progenitors in mouse models of AML

ABSTRACT: This is a RNA-Seq study from paired mutant-wild type mouse haemopoietic cells (n=4 mutant strains with 6 mice per strain - 3 mutant & 3 wild type). We want to see the transcriptomic effects of these mutation on blood progenitors.

Project description:The Arabidopsis Branching Enzyme 1 (BE1) gene encodes a putative glycoside hydrolase involved in carbohydrate metabolism. A partial loss-of-function mutation of the BE1 gene (be1-3 mutant) severely impaired adventitious shoot formation and somatic embryogenesis but not root formation in tissue culture. To gain a better understanding of the molecular mechanism underlying the in vitro plant regeneration defects caused by the BE1 gene mutation, we performed RNA sequencing analysis (RNA-seq) to examine the differential gene expression between WS and be1-3 mutant at dedifferentiation and redifferentiation stages.

Project description:The pistillody mutant wheat (Triticum aestivum L.) plant HTS-1 exhibits homeotic transformation of stamens into pistils or pistil-like structures. Unlike common wheat varieties, HTS-1 produces three to six pistils per floret, potentially increasing the yield. Thus, HTS-1 is highly valuable in the study of floral development in wheat. In this study, we conducted RNA sequencing of the transcriptomes of the pistillody stamen (PS) and the pistil (P) from HTS-1 plants, and the stamen (S) from the non-pistillody control variety Chinese Spring TP to gain insights into pistil and stamen development in wheat.

Project description:Arabidopsis thaliana infiltrated with different strains of DC3000 were used to elicit PTI, ETI, or a disease response compared to a mock control. This experiment was carried out in both Col-0 wild-type plants as well as mutants deficient in non-sense mediated decay via disruption of the promoter of upf1 (genotype background upf1-5). To identify differentially expressed and alternatively spliced transcripts elicited by bacterial infection, the NMD-impaired and wild-type host transcriptomes were profiled using RNA-seq. Wild-type and upf1-5 mutant plants were independently challenged with pathogenic (DC3000 COR-) or non-pathogenic P. syringae strains (DC3000 COR- ÎhrpS and DC3000 COR- avrPphB).

Project description:To gain initial insight into the regulatory mechanisms by which signalling in response to an elevated CO2 concentration exerts CA1- and CA4-dependent repression of stomatal development, we conducted high-throughput RNA-seq transcriptomics on immature aerial tissues of A. thaliana seedlings grown at the low (150 p.p.m) and elevated CO2 concentrations (500 p.p.m). Hypocotyls and cotyledons of developing seedlings (5 DAG; WT and ca1 ca4 mutant plants; n>1,000 per sample) grown in the low and elevated CO2 concentrations were used as source tissue to extract total RNA and conduct RNA-seq experiments using the HiSeq 2000 platform (Illumina). Transcriptome of bhlh010, bhlh089 and bhlh091 mutant anthers

Project description:Transcriptomic analysis of single, double and triple mutant anthers of bhlh010, bhlh089 and bhlh091. We examine here three recently duplicated Arabidopsis bHLH genes, bHLH010, bHLH089 and bHLH091, using evolutionary, genetic, morphological and transcriptomic approaches, and uncover their redundant functions in anther development. These three genes are relatively highly expressed in the tapetum of the Arabidopsis anther; single mutants at each of the bHLH010, bHLH089 and bHLH091 loci are developmentally normal, but the various double and triple combinations progressively exhibit increasingly defective anther phenotypes (abnormal tapetum morphology, delayed callose degeneration, and aborted pollen development), indicating their redundant functions in male fertility. Note: Samples in SRA were assigned the same sample accession. This is incorrect as there are different samples, hence “Source Name” was replaced with new values. Comment[ENA_SAMPLE] contains the original SRA sample accessions.
