Transcription profiling of human CD14+ monocytes treated with innate defence regulator peptide IDR-1 in the presence/absence of lipopolyssacharide

ABSTRACT: Effect of peptide IDR-1 on CD14+ human monocytes. >br< >br< Effect of peptide IDR-1 on LPS-induced responses in CD14+ human monocytes.>br< >br< Human peripheral blood mononuclear cells (PBMC) were treated with innate defence regulator peptide IDR-1 in the presence and absence of LPS (purified from Pseudomonas aeruginosa) for 4hrs. The objective was to obtain a transcriptional profile of responses induced by IDR-1 and the effect of IDR-1 on LPS-induced transcription.

We show that an innate defense-regulator peptide (IDR-1) was protective in mouse models of infection with important Gram-positive and Gram-negative pathogens, including methicillin-resistant Staphylococcus aureus, vancomycin-resistant Enterococcus and Salmonella enterica serovar Typhimurium. When given from 48 h before to 6 h after infection, the peptide was effective by both local and systemic administration. Because protection by IDR-1 was prevented by in vivo depletion of monocytes and macrop  ...[more]
