Expression data from WT (Columbia) and pifq (pif1pif3pif4pif5) mutant Arabidopsis seedlings

Project description:Plants respond to changes in the red:far red ratio (R:FR) of incident light. A reduction in this ratio (increase in FR) results in the Shade Avoidance Response (SAR) with associated changes in gene expression. The Phyotchrome-Interacting Factors (PIFs) are bHLH transcription factors known to be involved in the SAR. An analysis of changes in gene expression in WT and quadruple pif1pif3pif4pif5 (pifq; Leivar et al., 2008 (PMID 19920208)) mutant seedlings in response to an increase in FR should identify primary targets of PIF signaling. We used microarrays to examine the SAR in WT (Columbia) and pifq mutant Arabidopsis seedlings. Arabidopsis WT and pifq mutant seeds were plated on GM medium without sucrose at room temperature. During this procedure, the seeds were routinely exposed to white light (WL) for a total of 1.5 hours after imbibition. Seeds were then stratified for 5 days at 4ºC in darkness, and then grown in WL (19 umol/m2/s, R/FR ratio of 6.48) for 2 days at 21°C (WL0 samples). Two-day-old WL-grown seedlings were then maintained in the same fluence rate of WL supplemented with far-red light (WL-FR, R/FR ratio of 0.006) for 1 (FR1), 3 (FR3) or 24 (FR24) hours before harvesting. Control seedlings were also maintained in parallel in the same fluence rate of WL for 24h (WL24) before harvesting. Three different biological replicates of each treatment were grown separately and extracted, processed, and analyzed independently.

Appressorial proteomics analysis of the wild-type and GAG biosynthetic mutant of Metarhizium robertsii

Project description:Exopolysaccharide galactosaminogalactan (GAG) is a fungal cell wall component composed of α-1,4 linked galactose, N-acetyl galactosamine and galactosamine, which has been demonstrated in Aspergillus fumigatus in association with fungal adhesion, biofilm formation and virulence. The gene cluster responsible for GAG biosynthesis has only been characterized in Aspergillus fungi. We found that the highly conserved gene cluster for GAG biosynthesis is also present in the insect pathogenic fungi Metarhizium species. Functional investigations in M. robertsii revealed that GAG is only produced on fungal cell wall during fungal germination, filamentation and the formation of the infection structure appressoria. Gene deletions revealed that, relative to the wild-type (WT), the appressorial mucilage production was abolished in the null mutant of M. robertsii. Since multiple enzymes are produced in appressorial mucilages, appressorial samples of the WT and mutant formed on cicada wings were collected and subjected to iTRAQ comparative proteomic analysis. We found that different protein families were up- or down-regulated in the null mutant when compared with the WT.

CAMTA3 is a key regulator of the general stress response, and is guarded by Dominant Suppressor of CAMTA3 1 and DSC2. Here we assay transcriptional response to elf18, an elicitor, at 10 (primary response), 30 (major GSR) and 90 (beginning to be immunity-specific responses) minutes, in wt and in a line mutated in camta3/dsc1/dsc2, thus losing the GSR with only partial autoimmune activation.

Transcription profiling of Arabidopsis wild type and sta1-1 mutant plants

Project description:In plants, many gene transcripts are very unstable, which is important for the tight control of their temporal and spatial expression patterns. To identify cellular factors controlling the stability of unstable mRNAs in plants, we used luciferase imaging in Arabidopsis to isolate a recessive mutant, stabilized 1 (sta1), with enhanced stability of the normally unstable luciferase transcript. The sta1 mutation also causes the stabilization of some endogenous gene transcripts and has a range of developmental and stress response phenotypes. STA1 encodes a nuclear protein similar to the human U5 snRNP-associated 102-kDa protein and to the yeast pre-mRNA splicing factor Prp1p and Prp6p. STA1 expression is up-regulated by cold stress, and the sta1 mutant is defective in the splicing of the cold-induced COR15A gene. Our results show that STA1 is a pre-mRNA splicing factor required for not only splicing but also the turnover of unstable transcripts and that it has an important role in plant responses to abiotic stresses. Experiment Overall Design: Five replicates for WT (Control) and two replicates for sta1 (mutant) were compared.

Project description:L. japonicus (Regel) Larsen cv, Gifu were initially obtained from Prof. Jens Stougaard (University Aarhus, Denmark) and then self-propagated at the University of Seville. Ljgln2-2 mutant was isolated from photorespiratory mutant screening as described previously (Orea et al., 2002; Mï¿½rquez et al., 2005; Betti et al., 2006). The mutant progeny of two consecutive back-crosses with WT was used for the present work. WT and mutant seeds were scarified and surface-sterilized, germinated in 1% agar Petri dishes, and transferred to pots using a 1:1 (v/v) mixture of vermiculite and sand as solid support. Five seedlings were planted in each pot and grown during 35 days in a growth chamber under 16/8 hours day/night, 20/18ï¿½C, with a photosynthetic photon flux density of 250 ï¿½mol/m2ï¿½s and a constant humidity of 70%. CO2 was automatically injected to a final concentration of 0.7% (v/v), to allow for normal growth of Ljgln2-2 mutant in a photorespiration suppressed atmosphere. Plants were watered with Hornum nutrient solution (Handberg & Stougaard, 1992). Drought was applied withholding irrigation for the reported period of time and sample plants or leaves were harvested for further analysis.
