Transcription profiling of Drosophila third instar larvae hemocytes TO EXPLAIN hemocyte development.

ABSTRACT: We measured gene expression profiles of third instar larvae hemocytes from selected strains related to hemocyte development. Every strain was purchased from commercial companies. And we used microarray data to find what biological processes show abnormalities first, and which gene expressions show similarities and differences between selected strains. Experiment Overall Design: 200~300 Drosophila third instar larvae hemocytes from each of five lines were dissected to get at least 1 microgram of total RNA. Because all samples were hybridized to single-dye Affymetrix chip, control data were compared with data from each mutant.

The gene expression profiles in response to fungal infection in hemocytes.

Project description:The object of the microarray data is to find several genes effected by fungal infection. In order to establish the gene expressino levels in response to fungal infection in hemocytes, B.bassiana were injected into the posterior end of third instar larvae using a tungsten needle and incubated for 1h at 25 celcius degree. To compare with non-infected hemocytes, total RNA from infected hemocytes were labeled by Cy5, total RNA from non-infected hemocytes were labeled by Cy3, and hybridized to chip.

Project description:Samples 1-8: Tissue-specific RNA sequencing (Illumina) using dissected ring glands isolated from TWO different time points of control (phm>w1118) third instar larvae. Time points are: light phase zt0-4 (which corresponde to 2-4 hours from second to third instar larvae molt); and dark phase zt18-22 (which corresponde to 16-20 hours from second to third instar larvae molt) Samples 9-32: Tissue-specific gene expression (RNA seq Illumina) using dissected ring glands isolated from TWO different time points of third instar larvae. Transcriptomic analysis of Drosophilalarval crystal cells

Project description:Crystal cells are one of the 3 Drosophila blood cell lineages and represent less than 5% of the total hemocytes in wild type larvae. There development is notably controlled by mlf (myeloid leukemia factor), which regulate their number by stabilising the lineage-specific transcription factor Lozenge. To gain insight into the biology of this blood cell lineage and its regulation by mlf, we established the gene expression profile of the circulating crystal cells in wildtype and mlf mutant third instar larvae. This study provides a rich source of information to further characterise crystal cell function and regulation. In addition our data show that mlf is a major regulator of crystal cell gene expression programm and that mlf mutation leads to the accumulation of misdifferentiated crystal cells.
