Identification of spondylarthropathy-specific genes in BALB/c mice with proteoglycan-induced spondylitis (PGIS)

ABSTRACT: BALB/c mice develop peripheral arthritis (PGIA) and spondyloarthropathy (PGIS) upon repeated intraperitoneal injections of human cartilage proteoglycan (PG) aggrecan. The aim of the present study was to identify spondylitis-specific genes by comparing intervertebral disc (IVD) RNA samples from spondyloarthropathic and naM-CM-/ve BALB/c mice. Keywords: Genetic modification We compared the gene expression data of 5 spondyloarthropathic and 3 naM-CM-/ve (control) mice.

Gene Expression Profile of Motheaten Mice (meb2) with novel mutation in Ptpn6 gene.

Project description:Gene expression array utilizes the advantage of the big number of the significantly changed genes creating transcriptional profiles of known mutations. Creating gene expression profiles of the animal models with known genetic defect provides great opportunity for further investigation human diseases with unknown etiology. Motheaten mice with mutation in protein tyrosine phosphatase Ptpn6 have massive neutrophilic infiltration within the skin followed by autoinflammatory and autoimmune systemic pathology resembling similarity to neutrophilic dermatoses in human. Keywords: genetic modification We performed microarray study on motheaten mice with novel mutation (meb2) in protein tyrosine phosphatase non-receptor-6 (Ptpn6) in with 5 sick bone marrow samples and 5 wild type controls and found significant difference between these two groups.

Regulatory mechanisms of TSG-6 (TNF-alfa-induced protein-6: Tnfip6)-deficient and wild-type synovial fibroblasts

Project description:BALB/c mice are susceptible to proteoglycan (PG) aggrecan-induced arthritis (PGIA), and the absence of TSG-6 further increases susceptibility and local inflammatory reactions, including neutrophil invasion into the joints. To gain insight into the mechanisms of TSG-6 action, synovial fibroblasts were isolated from wild-type and TSG-6-KO mice, cultured and exposed to various agents affecting either the TSG-6 expression and/or modify the intracellular function of TSG-6. In the present microarray studies, we have identified differences in gene expression by fibroblasts isolated from wild-type or TSG-6-KO mice Keywords: Genetic modification In this study we compared the gene expression profile of 42 fibroblast cultures from mouse synovial samples. Fifteen samples were wild-type and 27 samples were TSG-6 knockout. In these two genotype groups fibroblasts were treated with TNFa, LPS, TSG-6, HA, or HA+TSG-6, and were compared to untreated samples in the same genotype group. We also compared the same treatments between KO and WT.

Gene expression profile of preclinical arthritis and its modulation by antigen-induced tolerance

Project description:During the course of adjuvant arthritis, maximal changes in gene expression were observed at the incubation phase. A major group of genes affected was related to immune activity. Tolerance induction by mycobacterial heat-shock protein 65 (Bhsp65), the disease-related antigen, caused upregulation of a large number of genes. These included immune activity genes as well as cell proliferation-related genes. We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated genes during this process. The draining lymph node cells (LNC) were harvested from arthritic rats at different phases of adjuvant arthritis (incubation,peak and recovery). Also tested were LNC of naïve rats. LNC were tested ex vivo. Total RNA isolated from LNC was used for testing with affymetrix gene chip following manufacturer's instructions. Similarly, LNC of Bhsp65-tolerized rats harvested at incubation phase were tested with or without Bhsp65 restimulation in vitro. LNC of arthritic rats in incubation phase were similarly restimulated with Bhsp65 in vitro as controls, and the results compared with that of the tolerized rats.

Periodontal pathogens do not exacerbate macroscopic arthritis but alter the synovial proteome

Project description:Rheumatoid arthritis (RA) and periodontitis (PD) are chronic inflammatory diseases that appear to occur in tandem. Autoantibodies against citrullinated peptide antigens linked pathogeneses with PD preceding RA. However, the mutual impact PD exerts on RA and vice versa has not yet been defined. To address this issue, we set up an animal model and analyzed how the prime inducers of citrullination - Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans ÃƒÂ¢Ã¢â€šÂ¬Ã¢â‚¬Å“ differ in their pathogenic potential. Our experimental setup included collagen induced arthritis (CIA) in the mouse, oral inoculation with P. gingivalis or A. actinomycetemcomitans to induce alveolar bone loss and the combination of both diseases in inverted orders of events. Label-free quantitative proteome analysis revealed that P. gingivalis and A. actinomycetemcomitans led to differential expression patterns in the synovial membranes that were reminiscent of cellular and humoral immune responses, respectively. The P. gingivalis and A. actinomycetemcomitans specific signatures in the synovial proteomes suggest a role for oral pathogens in shaping disease subtypes and setting the stage for subsequent therapy response.

An integrated RNA-Seq and network study reveals the effect of nicotinamide on adrenal androgen synthesis

Project description:Acne vulgaris is a chronic inflammatory disease of the skin resulting from androgeninduced increased sebum production and altered keratinization. Nicotinamide (NAM), an amide form of vitamin B3 with a well-established safety profile, has shown good therapeutic potential in treating acne and its complications. NAM has anti-inflammatory effects and reduces sebum but its function in androgen biosynthesis remains unknown. In this study, we used a widely used cell model, starved human adrenal NCI-H295R cells, to examine the effects of NAM in androgen production and its mediated network changes. By treating NCI-H295R cells with 1-25 mmol/L of NAM, we found that cell viability was only slightly inhibited at the highest dose (25 mmol/L). NAM reduced testosterone production in a dose-dependent manner. Transcriptomic analysis demonstrated that key enzymes of androgen biosynthesis were significantly decreased under NAM treatment. In addition, gene set enrichment analysis (GSEA) showed that gene sets of cell cycle, steroid biosynthesis, TGFβ signalling, and targets of IGF1 or IGF2 were enriched in NAM-treated cells. Kyoto Encyclopaedia of Genes and Genomes (KEGG) pathway and Gene ontology (GO) analysis of the differentially expressed genes also suggested that steroidogenesis and SMAD signalling were affected by NAM. Overall, these crucial genes and pathways might form a complex network in NAM-treated NCI-H295R cells and result in androgen reduction. These findings help explain the potential molecular actions of NAM in acne vulgaris, and position NAM as a candidate for the treatment of other hyperandrogenic disorders.

Transcription profiling of BALB/c mice genetically susceptible to proteoglycan-induced arthritis and spondylitis

Project description:BALB/c mice are susceptible to proteoglycan (PG) aggrecan-induced arthritis (PGIA), a murine model of rheumatoid arthritis (Glant,T.T. and Mikecz,K., Proteoglycan aggrecan-induced arthritis. A murine autoimmune model of rheumatoid arthritis. Methods Mol.Med. 2004. 102: 313-338.). However, there are marked differences among BALB/c colonies (maintained by different vendors at different locations) in PGIA onset and severity, which could be the result of subtle variations in their genetic background. In the present microarray study, we have identified differences among BALB/c colonies, and an altered immunization-related gene expression pattern in PGIA model. Experiment Overall Design: In this study we compared the gene expression profile of 12 spleens from PG-immunized (RNA was isolated 12 days after immunization) and naive mice from BALB/cJ and BALB/cByJ colonies (3-3 from each group), then made comparisons between colonies and based on immunization.

Project description:Prednisolone is a potent anti-inflammatory glucocorticoid (GC) but chronic use is hampered by metabolic side effects. Although GCs are predominantly prescribed for treatment of inflammatory conditions, little is known about their long-term effects on gene-expression in-vivo during inflammation. Here, we aimed to identify genes underlying prednisolone-induced metabolic side effects in a complex in-vivo inflammatory setting after long-term treatment. We performed whole-genome expression profiling in liver and muscle from arthritic and non-arthritic mice treated with several doses of prednisolone for three weeks.
