Expression data from LCM-captured Zea mays root cells after challenge with Meloidogyne incognita

Project description:The differentiation of specialized feeding sites in Zea mays root cells in response to nematode infestation involves substantial cellular reprogramming of host cells that is not well characterized at the molecular level. Expression data was generated from Zea mays root cells undergoing giant cell formation due to nematode infestation and from non-infested control root cells. Cells were laser captured 14 and 21 days after infestation. Each time point (14 day and 21 day) consisted of three biological replicates per treatment (control root cells or giant cells). Control cells were captured from an area ~13,000,000 um2 in size and giant cells were captured from an area ~5,000,000 um2 in size. RNA samples were isolated using the PicoPure RNA Isolation Kit (Arcturus, Mountain View, USA). RNA amplifications were carried out with the NuGEN WT-Ovation Pico kit.

Proteomic Analysis Reveals the Major Proteins and Pathways that Mediate the Effect of 17-β-Estradiol in Cell Division and Apoptosis in Breast Cancer MCF7 Cells

Project description:Despite extensive research, the genes/proteins and biological pathways responsible for the physiological effects of estrogen remain elusive. In this study, we employed a proteomic method to determine the effect of estrogen on global protein expression in breast cancer MCF7 cells. The expression of 77 cytoplasmic, 74 nuclear, and 81 membrane proteins was significantly altered by 17-β-estradiol (E2). Protein enrichment analyses of the quantified proteins revealed that E2 stimulated cell division primarily by promoting the G1 to S phase transition and advancing the G2/M checkpoint. Functional annotation analyses of the E2-regulated proteins indicated that these proteins promoted cell division primarily through enhancing protein nuclear import, protein translation, reducing mRNA degradation, and increasing protein folding. Interestingly, many of the E2-upregulated proteins contained the HEAT, KH, and RRM domains. The effect of E2 on cell survival was complex, as it could simultaneously enhance and inhibit apoptosis. E2 enhanced apoptosis by promoting cytochrome c release from mitochondria and inhibited apoptosis by activating the PI3K/AKT/mTOR signaling pathway. Treatment of MCF7 cells with E2 and the PI3K inhibitor Ly294002 significantly enhanced apoptosis compared to the cells treated with E2 alone. Our results suggest that combining estrogen with a PI3K inhibitor could be a promising strategy for treating ERα-positive breast cancer.

Dietary sterols/steroids and the generalist caterpillar Helicoverpa zea: physiology, biochemistry and midgut gene expression

Project description:Sterols are essential nutrients for insects because, in contrast to mammals, no insect (or arthropod for that matter) can synthesize sterols de novo. Cholesterol is the most common sterol in insects, but it is not found in plants in large quantities; plant-feeding insects typically generate their cholesterol by metabolizing phytosterols. However, different plants species can contain different types of phytosterols, and some phytosterols are not readily converted to cholesterol. In this study we examined, using artificial diets containing single sterols, how typical (cholesterol and stigmasterol) and atypical (cholestanol and cholestanone) sterols/steroids affect the performance of a generalist caterpillar (Helicoverpa zea), restricting this analysis to midgut tissue because this is where sterol/steroid absorption occurs, and the midgut is the putative site of dietary sterol/steroid metabolism. In general, H. zea performed best on the cholesterol and stigmasterol treatments; performance was reduced on cholestanol, and was very poor on cholestanone. We compared the transcript profiles of larval guts in response to differentially suitable sterols, using the optimal sterol, cholesterol, as a control, using a two-color reference design microarray experiment. Midgut gene expression patterns differed between the treatments; relative to cholesterol, differences were lowest on the stigmasterol treatment, intermediate on the cholestanol treatment, and greatest on the cholestanone treatment. Early transcriptional changes in Zea mays seedlings exposed to Z-3-hexenol

Project description:Z-3-Hexenol and other green leaf volatiles have been known to induce defense-related gene expression. Here we investigated the early transcriptional changes in response to Z-3-hexenol. 3-week-old Zea mays seedling (inbred line B73) was exposed to pure Z-3-hexenol (1.5 Î¼M) in a glass cylinder. Controls were treated likewise without the addition of Z-3-hexenol. Plants were exposed for 20 min and 60 min. The second leaf was then cut and shock-frozen in liquid N2 and then stored at -85Â°prior to RNA extraction. 3 individual leaves were pooled for 1 biological replicate. 3 biological replicates were performed for each treatment, with one dye swap (second sample).

Transcriptome sequencing of and microarray development for a Helicoverpa zea cell line to investigate in vitro insect cell-baculovirus interactions

Project description:The Helicoverpa armigera single-capsid nucleopolyhedrovirus (HaSNPV) can be propagated using H. zea insect cell cultures, for use as a biopesticide against Heliothine agricultural pests.This study sequenced, assembled and functionally annotated 29,586 transcript sequences from cultured H. zea cells using Illumina 100 bps and paired-end transcriptome sequencing (RNA-seq). From these sequences, a genome-scale microarray platform was constructed and validated for effective expression analysis of H. zea genes. This array also included probes for all HaSNPV genes, thereby allowing virus and host gene changes to be monitored simultaneously. A 4x180,000 SurePrint Agilent expression array (Agilent, Santa Clara, CA) was employed so that a high number of probes can be included to test all 29,586 assembled H. zea sequences and to eventually select a best probe for each transcript. Two biological replicates for uninfected H. zea cells and two other biological replicates for infected samples at 18 hours post infection were analyzed. Six Agilent 60-mer oligonucleotide probes were designed by eArray (Agilent) for each transcript, in which each orientation had three probes randomly distributed across the sequences. Probes that had potential cross-hybridization (Xhyb) were removed. The final probe set for H. zea sequences included 153,583 probes. In addition, probes for all 135 H.armigera single-capsid nucleopolyhedrovirus (HaSNPV) genes were added to investigate host-virus interaction in culture.
