Project description:White mold disease, caused by Sclerotinia sclerotiorum (Lib) de Bary, can be a serious disease of crops grown under cool moist environments. In many plants, like soybean [Glycine max (L.) Merr.], complete genetic resistance does not exist. To identify possible genes involved in defense against this pathogen, and to determine possible physiological changes that occur during infection, a microarray screen was conducted using stem tissue to evaluate changes in gene expression between partially resistant and susceptible soybean genotypes at 8 and 14 hours post inoculation. RNA from 15 day old inoculated plants was labeled and hybridized to soybean cDNA microarrays. ANOVA identified 1270 significant genes from the comparison between time points and 105 genes from the comparison between genotypes. Selected genes were classified into functional categories. The analyses identified changes in cell-wall composition and signaling pathways, as well as suggesting a role for anthocyanin and anthocianidin synthesis in the defense against S. sclerotiorum. In-silico mapping of both the differentially expressed transcripts and of public markers associated with partial resistance to white mold, provided evidence of several differentially expressed genes being closely positioned to white mold resistance markers, with the two most promising genes encoding a PR-5 and anthocyanidin synthase. Project description:Legumes interact with nodulating bacteria that convert atmospheric nitrogen into ammonia for plant use. This nitrogen fixation takes place within root nodules that form after infection of root hairs by compatible rhizobia. Using cDNA microarrays, we monitored gene expression in soybean (Glycine max) inoculated with the nodulating bacterium Bradyrhizobium japonicum 4, 8, and 16 days after inoculation (dai), time points that coincided with nodule development and the onset of nitrogen fixation. This experiment identified several thousand genes that were differentially expressed in response to B. japonicum inoculation. Expression of 27 genes was analyzed by qRT-PCR and their expression patterns mimicked the microarray results confirming integrity of analyses. The microarray results suggest that B. japonicum reduces plant defense responses during nodule development. In addition, the data revealed a high level of regulatory complexity (transcriptional, post-transcriptional, translational, post-translational) that is likely essential for development of the symbiosis and adjustment to an altered nutritional status. Project description:Atrazine and bentazon are both photosystem II inhibiting herbicides that interfere with photosynthetic electron transport provoking oxidative stress. While atrazine is lethal to soybean, bentazon does not kill soybeans because of the capability of soybeans to metabolize the herbicide. Gene expression profiling was conducted using cDNA microarrays to understand the responses of soybeans to PSII interruption and concomitant oxidative stress caused by atrazine and bentazon by monitoring expression at 1, 2, 4, and 8 h after treatment (HAT). The microarray study revealed that 6,646 genes were differentially expressed with high statistical significance over the experiment with 88% of them sharing similar expression pattern between the atrazine and bentazon treatments. Many genes related to xenobiotic detoxification and antioxidation such as cytochrome P450s, glutathione-S-transferases, superoxide dismutases, catalases and tocopheral cyclases were induced by the herbicides. The study also discovered plants treated with bentazon started to recover between 4 HAT and 8 HAT as reflected in the decreased amplitude of fold changes of most genes from 4 to 8 HAT. The 12% of the genes that were differentially expressed between atrazine and bentazon were largely related to cell recovery, such as genes related to ribosomal components. Project description:This SuperSeries is composed of the following subset Series: GSE13254: Sclerotinia infected vs Mock infected controls in B. napus (Westar) GSE13256: Zhong You 821 Sclerotinia infected vs Mock infected controls in B. napus (Zhong You 821) The fungal pathogen Sclerotinia sclerotiorum infects a broad range of dicotyledonous plant species and is the causative agent of stem rot in Brassica napus. To elucidate the mechanisms underlying the defense response, we studied the patterns of gene expression in a partially resistant variety of ZhongYou 821 (ZY821) and a susceptible line from Westar over five time points, 6, 12, 24, 48, and 72 hours post-inoculation (hpi) using a B. napus oligonucleotide microarray. For each cultivar, a two-dye experiment was run comparing infected to mock-infected stem tissue. For each time point, 6 microarray slides were done (3 biological replicates, with a dye swap for each biological replicate). Refer to individual Series
