Genome wide rhythmic transcription under light/dark cycles in Ostreococcus tauri

ABSTRACT: Genome wide rhythmic transcription under light/dark cycles is associated with sequential transcription of specific biological processes genes in Ostreococcus tauri. Transcriptional profiling of Ostreococcus tauri under light/dark cycles. In order to identify genes with a diurnal rhythm, cells entrained under 12:12 light/dark cycles were sampled every 3 hours for 27 hours with two overlapping time points at Time 9 (Light ON at Time 9; Light OFF at Time 21) in 3 independent experiments. Two-conditions experiment, test situation (kinetic point) vs. Pool of each samples. Biological triplicates: 3 independant kinetics, 9 points per kinetics, Technical triplicates per array.

Project description:The overall set of experiments consist of sampling a culture of the unicellular algae Ostreococcus tauri (wild type) every 24 hours for 4 time points(ZT24, ZT48, ZT72 and ZT96). Each time point was performed with 5 replicate culture flasks, the cultures were grown under dark conditions. This proteome is associated to the transition from an 12:12 light:dark to a dark:dark regime.

Unveiling Seasonal Adaptive Molecular Mechanisms in Marine Phytoplankton during Diel Cycles through Multiomics Integration

Project description:Earth tilted rotation and translation around the Sun produce one of the most pervasive rhythms on our planet, giving rise to seasonal variations in diel cycles. Although marine phytoplankton plays a key role on ecosystems, multiomics analysis of its response to these periodic environmental signals remains largely unexplored. The marine picoalga Ostreococcus tauri, which resides at the base of the green lineage, has been chosen as model organism due to its high abundance in marine phytoplankton and its cellular and genomic simplicity, making it the smallest free living eukaryote. Ostreococcus was subjected to different light regimes: summer long days, winter short days, constant light and constant dark conditions, to investigate these responses. Although 80% of the transcriptome presented diel rhythmicity, less than 5% maintained oscillations under constant conditions. A drastic reduction in proteome rhythmicity was observed with 55% of the proteins oscillating. Seasonal specific rhythms were found for key physiological processes such as cell cycle, photosynthesis, carotenoid biosynthesis, starch accumulation and nitrate assimilation. In this study, a global orchestration between transcriptome, proteome and physiological dynamics was characterised identifying specific seasonal temporal offsets between peaks in transcripts, proteins, and physiological responses.

Project description:Transcript profiling was performed on samples derived from plants grown under long day conditions. Time series were harvested under diurnal (L/D; light/dark cycles) at control temperature (20 C) and cold (4 C); and under circadian conditions (L/L; continuous light) at control temperature (20 C). Leaves were sampled at time 0 and after 2 h (to coincide with light-dark transitions) and then every 4 h until 58 h.

Nocturnal expression data of Synechococcus in WT and kaiABC-null strains

Project description:In the unicellular cyanobacterium, Synechococcus elongatus PCC 7942, most of genes are downregulated in the dark, while 10% of genes are upregulated. Here, we employed high-density oligonucleotide arrays to explore comprehensive profiles of genome-wide Synechococcus gene expression in wild type and kaiABC-null strains under continuous dark (DD) conditions.
