ABSTRACT: Global expression profile of human osteoblast treated with chemotherapy-treated bone marrow stromal cell conditioned media, compared to human osteoblast cells treated with diluent-control bone marrow stromal cell conditioned media. Goal is to identify genes regulated by chemotherapy in osteoblasts. Dye-swap design with 6 biological replicates. Three arrays performed with chemotherapy treated samples on channel 1 and diluent-control treated on channel 2; three arrays performed with chemotherapy treated samples on channel 2 and diluent-control on channel 1.

Project description:Purpose: We have used microarrays to identify gene expression profiles that distinguish mouse OS cells from normal pre-osteoblast cells and mature osteoblast cells. Methods: Transcriptional profiles of three cell lines derived from tumors from Osx-Cre p53fl/fl Rbfl/fl (fibroblastic OS) mouse model, and from pre-osteoblast cells (Kusa4b10 mouse bone marrow stromal cell line) and osteoblast cells (derived by in vitro differentiation of the Kusab410 mouse bone marrow stromal cell line) were generated by microarray analysis, each in triplicate, using Affymetrix mouse Gene1.0ST arrays. Transcriptional profiles were analyzed in cell lines derived from tumors from a genetically engineered mouse model of human osteosarcoma (Osx-Cre p53fl/fl Rbfl/fl) and osteoblast cells derived from the Kusa4b10 mouse bone marrow stromal cell line, in the undifferentiated state (pre-osteoblasts) and differentiated state (osteoblasts).

Global gene expression analysis of heterotypic interactions between cancer cells and osteoblasts in vitro

Project description:Bone is the most common site of breast cancer metastasis, and this type of metastasis is a main cause of morbidity. Because breast cancer is a heterogeneous disease, the interactions between the cancer cells and the skeletal host cells, such as osteoblasts, might be diverse. Thus far, these tumor-osteoblast interactions have not yet been well characterized using a genomic approach. We hypothesized that gene expression signatures induced by tumor-osteoblast interactions might be of clinical relevance. To examine these gene expression signatures, we established an ex vivo co-culture model with benign human cells and a panel of 5 malignant breast epithelial cells in combination with primary human osteoblasts and determined associated gene expression changes using cDNA microarrays. Project description:This study is designed to identify gene promoters and enhancers target of foxo1, an transcription factor activated by melatonin in osteoblast cells. Protocol:1. Primary osteoblast cells were grown at 70-80% confluency in 150mm culture dishes.2. These cells were then treated with melatonin for 24h under serum-deprived condition.3. After the completion of treatment cells were fixed with medium containing formaldehyde, washed with PBS and reaction was stopped by adding glycine stop solution. Cells were collected in cell scraping solution. 4. Further sonication and CHIP reaction with foxo1 antibody was performed using Active Motif CHIP-IT express kit following manufacturers instructions.5.
