ABSTRACT: affy_meja_arabidopsis - We want to determine if the presence of C2 modifies the cell transcriptome and its response to methyl jasmonate. Comparison of control lines and transgenic lines expressing C2 in basal conditions and after treatment with 50uM MeJA for 10 hours. C2 lines are expressing geminivirus C2 under a 35S promoter. Elf18-induced long noncoding RNA associates with Mediator to enhance expression of innate immune response genes in Arabidopsis

Project description:Plant immune response is a complex process involving both transcriptional and posttranscriptional regulation of gene expression. Here, we identify an Arabidopsis long-noncoding (lnc) RNA, designated elf18-induced long noncoding RNA 1 (ELENA1), as a factor enhancing resistance against Pseudomonas syringe pv. tomato DC3000. ELENA1 knock-down plants show a decreased expression of PR1 and the plants are susceptible to pathogens. By contrast, plants over-expressing ELENA1 show elevated Pathogenesis-Related gene 1 (PR1) expression after elf18 treatment and display pathogen resistance phenotype. RNA-seq analysis of ELENA1 overexpressing plants after elf18 treatment confirms increased expression of defense-related genes compared to WT. ELENA1 directly interacts with Mediator subunit 19a (MED19a) and affects enrichment of MED19a on PR1 promoter. These results show that ELENA1 is a novel lncRNA that regulates PR1 expression through MED19a. Our findings uncover an additional layer of complexity implicating lncRNA in the transcriptional regulation of plant innate immunity.

Project description:This study has examined the molecular mechanisms underlying sensitivity of sarcomas to Nutlin-3a, a non-genotoxic activator of the p53 pathway. Human patient material was collected immediately following surgical resection, dissected into small pieces and ex planted onto gelatin sponges immersed in media containing either vehicle control or Nutlin-3a (10uM and/or 50uM) for 48 hours. Cephalotaxus hainanensis LC-MSMS

Project description:In previous work, cephalotaxine, harringtonine, homoharringtonine were shown to be accumulated differentially after various stimuli. Especially, after MeJA treatment, the concentration of 3 cephalotaxus alkaloids all showed decreasing. We speculated that the genes expressed lower after MeJA treatment might encode some enzymes responsible for the biosynthesis of cephalotaxus alkaloids. Therefore, choosing the sample treated with MeJA and the control sample for comparative iTRAQ analysis will greatly facilitate dissection of the genes involved in the biosynthesis of cephalotaxus alkaloids and even the acyl portions of cephalotaxus ester alkaloids. This approach is widely used for mining and identifying novel genes in the biosynthesis of secondary metabolites without genome data in plants.
