Genome wide transcriptional profiling of c-Cbl deficient hematopoietic stem and progenitor cells

ABSTRACT: Expression of genes that are differentially expressed between c-Cbl control and c-Cbl mutant hematopoietic stem and progenitor cells were identified through this approach. Interestingly, several leukemia associated genes, particularly transcription factors, were differentially expressed between c-Cbl WT and c-Cbl mutant hematopoietic stem and progenitor cells. Hematopoietic stem and progenitor cells of c-Cbl control and c-Cbl mutant mice were sorted based on their defined immunophenotype. Gene expression studies were performed using Illumina microarray chips.

MicroRNAs and Metabolites in Serum Change after Chemotherapy and Impact on Haematopoietic Progenitor Cells

Project description:Hematopoietic reconstitution after chemotherapy and hematopoietic stem cell transplantation (HSCT) requires massive activation of the stem cell pool. In a previous study, we have shown that there are systemically released factors in patient serum after HSCT that may play an important role in hematopoietic recovery, and that these serum samples significantly stimulate hematopoietic stem and progenitor cell (HPC) in vitro expansion. Aberrant microRNA (miRNA) expression is a feature of leukemia and miRNAs also play a significant role in normal hematopoiesis and differentiation. In this study, we have addressed the question if miRNAs and metabolic pathways are involved in HPC activation after HSCT. Serum from patients before chemotherapy and after chemotherapy during aplasia was used as supplement for in vitro cultivation of CD34+ HPCs. Serum taken after chemotherapy significantly enhanced HSC proliferation and maintained a primitive CD34+ immunophenotype. Microarray analysis revealed that 23 miRNAs are differentially expressed in serum before and after chemotherapy - particularly miRNA-320 and miRNA-1275 were down-regulated and miRNA-3663-3p was up-regulated. This suggest that hematopoietic regeneration after chemotherapy might be tightly regulated by miRNAs and this opens new perspectives for in vivo stimulation of the stem cell pool.

Genome-wide comparison of the transcriptomes of highly enriched normal and chronic myeloid leukemia stem and progenitor cell populations

Project description:The resistance of CML leukemic stem cells (LSC) to tyrosine kinase inhibitor therapies targeting BCR-ABL leads to persistence of disease in most cases. We have identified novel putative therapeutic targets that are differentially expressed in CML LSCs compared to normal hematopoietic stem cells (HSC) by transciptional profiling of stem and progenitor cell populations from CML patients and normal donors. These data are used to obtain 97 genes that are differentially expressed in CML vs. normal stem and progenitor cells and 236 transcripts that show evidence of alternative exon usage in CML vs. normal stem cells. Bone marrow stem and progenitor cell fractions from 5 patients with newly diagnosed and untreated chronic phase CML and 5 healthy donors were analyzed. Contrasts between CML vs. normal stem and progenitors overall and CML vs. normal stem cells were performed using Partek Genomic Suite software. Genes with |log2(fold-change) > 1| and false discovery rate (FDR) of 0.05 were identified as significantly differentially expressed. To identify genes with evidence of alternative exon useage, contrasts between CML and normal stem cell populations were performed, and genes with an alternative splicing FDR of 0.01 were considered to be alternatively spliced.

Proteomic characterization of CD34+ CD123+ stem cells from patients with acute myeloid leukemia

Project description:The comparative characterization of hematopoietic stem cells from healthy stem cell donors and patients with acute myeloid leukemia on a proteome level has the potential to reveal differentially regulated proteins which might be candidates for specific immunotherapy target molecules. Exemplarily, we analyzed the proteome of the cytosolic and the membrane fraction of CD34 and CD123 co-expressing FACS-sorted leukemic progenitors from five patients with acute myeloid leukemia employing mass spectrometry. As a reference, CD34+CD123+ normal hematopoietic progenitor cells from five healthy stem cell donors were analyzed. In this TMT 10-plex labeling based approach 2068 proteins were identified with 256 proteins differentially regulated in one or both cellular compartments. This study demonstrates the feasibility of a mass spectrometry based proteomic approach to detect differentially expressed proteins in two compartment fractions of leukemic stem cells as compared to their healthy stem cell counterparts.

Transcription profiling of human hematopoietic stem/progenitor cells treated with TGF-beta1 to idetntify its target genes (reference series)

Project description:Leukemias and other cancers possess a rare population of cells capable of self-renewal, and eradication of these cancer stem cells is likely necessary for long-term cancer-free survival. Given that both normal and cancer stem cells are capable of self-renewal the extent to which cancer stem cells resemble normal tissue stem cells is a critical issue if targeted therapies are to be developed. We introduced the MLL-AF9 fusion protein encoded by the t(9;11)(p22;q23) found in human acute myelogenous leukemia (AML) into murine committed granulocyte-macrophage progenitors (GMP). The resultant leukemias contained cells with an immunophenotype similar to normal GMP that were highly enriched for leukemia stem cells (LSC). Detailed gene expression comparisons between normal hematopoietic stem cells (HSC), committed progenitors, and the LSC population demonstrated the LSC were globally more similar to the normal GMP than any other population. However, a subset of genes highly expressed in normal stem cells was re-activated in the LSC. These data demonstrate LSC can be generated from committed progenitors without widespread reprogramming of gene expression, and a leukemia self-renewal associated signature is activated in the process. Our findings define progression from normal hematopoietic progenitor to leukemia stem cell, and suggest that targeting a self-renewal program expressed in an abnormal context may be possible.
