Gene expression from SCID-hu mice treated with saline (control) or intermittent PTH

ABSTRACT: The data present global gene expression profile of whole human bones, implanted in SCID mice (SCID-hu model), then engrafted with the myeloma cell line, Hg, and treated with saline or PTH for 4 weeks. SCID-hu mice were prepared by subcutaneous implantation of human bone in SCID mice. The human myeloma cell line, Hg was injected directly into the implanted bone in SCID-hum mice. Upon establishment of myeloma growth as determined by measurement of circulating human immunoglobulins in SCID-hu mice sera, the mice were treated with saline or intermittent PTH (parathyroid hormone, 80 ug/kg/day) for 4 weeks. At the end of the experiment, the human bones from SCID-hu mice were snap frozen in liquid nitrogen. RNA extracted from these bones was subjected to global gene expression profile using Affymetrix U133-Plus microarray.

Inflammation driven by tumor-specific Th1 cells protects against B-cell cancer

Project description:The immune system can both promote and suppress cancer. Chronic inflammation and proinflammatory cytokines such as interleukin (IL)-1 and IL-6 are considered tumor-promoting. In contrast, the exact nature of protective antitumor immunity remains obscure. In this study, we have quantified locally secreted cytokines during primary immune responses against myeloma and B-cell lymphoma in mice. Strikingly, successful cancer immunosurveillance mediated by tumor-specific CD4+ T cells was consistently associated with elevated local levels of both proinflammatory (IL-1aplha, IL-1beta, and IL-6) and T helper 1 (Th1)-associated cytokines (interferon-alpha, IL-2, IL-12). Cancer eradication was achieved by a collaboration between tumor-specific Th1 cells and tumor-infiltrating, antigen-presenting macrophages. Th1 cells induced secretion of IL-1? and IL-6 by macrophages. Th1-derived interferon-? was shown to render macrophages directly cytotoxic to cancer cells, and to induce macrophages to secrete the angiostatic chemokines CXCL9/MIG and CXCL10/IP-10. Thus, inflammation, when driven by tumor-specific Th1 cells, may prevent rather than promote cancer. Tumoricidal macrophages were isolated from Idiotype-specific TCR-transgenic SCID mice injected with MOPC315-containing Matrigel. Control macrophages were obtained from TCR-transgenic SCID mice injected with Matrigel containing antigen-loss MOPC315.

Comparison of gene expression profiles by CD3+CD4+ thymocytes derived from fetal and adult hematopoietic stem cells

Project description:Human fetal and adult hematopoietic stem cells (HSC) were obtained from fetal liver, fetal bone marrow (BM), and adult BM. These were injected into human fetal thymic implants in SCID-hu Thy/Liv mice (4-6 separate mice per HSC donor) and allowed to mature into single positive CD4+ (SP4) thymocytes over the course of 7-8 weeks. SP4 thymocytes from injected stem cells were subsequently sort-purified from thymic implants and gene expression was performed. HSC from fetal (age 18-22 gestational weeks) and adult (age: 19-43 year old) HLA-A2+ donors were obtained from different tissues. After injection into human fetal thymic implants (SCID-hu Thy/Liv HLA-A2-) the cells were allowed to mature into thymocytes and sorted on the basis of HLA-A2+ expression and CD3+CD4+ (SP4) expression. 3 separate thymic implants were analyzed for each group.

Multinucleated Tartrate Resistant Acid Phosphatase Positive Cells with Macrophage and Osteoclast Phenotypes in Bone

Project description:Prothrombin (PT) and osteopontin (OPN) promotes adhesion of different TRAP-positive multinucleated cells isolated from rat long bone (Hu et al. Exp Cell Res. 2008; 314: 638-50). The PT-adhering cell could represent either an immature precursor to the OPN-adherent osteoclast or constitute a distinct multinucleated cell population with a unique role in bone. Herein, phenotypic differences between PT- and OPN- cells were investigated with microarray- expression analysis. Characteristic for PT-cells was expression of innate immune response genes and scavenger receptors whereas OPN-cells expressed typical osteoclast proteins such as collagenases implicated in bone degradation. Bone cells were extracted from neonatal Sprague Dawley rat long bones. The extracted cells were attached to PT or OPN, and thereby two cell populations were isolated. mRNA was extracted from these cells and their expression was compared with the Affymetrix GeneChip Rat Gene 1.0 ST Array.

Gene expression data from teratomas formed by human iPSCs (chiPS) in SCID (Severe combined immunodeficiency) mice and humanized SCID mice (HuSCID)

Project description:Human fetal liver fibroblast were reprogrammed to chiPS without exogenous DNA integration using a single episomal vector. The chiPS were then transplanted into SCID mice and HuSCID to form teratomas. We used microarrays to profile the gene expression differences between the teratomas formed by chiPS in SCID and HuSCID mice. Pooled total RNA from two teratomas formed by chiPS in SCID and HuSCID mice respectively were used for hybridization on Affymetrix microarrays.
