Affy_l2bctv_ath-Expression viral protein L2 BCTV in Arabidopsis thaliana

ABSTRACT: affy_l2bctv_ath - affy_l2bctv_ath - Geminivirus diseases are among the most important economic impact on crops, including beans, cassava, cotton, cucurbits, corn, peppers and tomatoes. Geminiviruses have a genome composed of one or two molecules of single-stranded DNA, depending on mono or bipartite. The BCTV genome consists of a single strand of DNA that contains six reading frames for proteins CP, V2, Rep, C2, Ren, and C4. To determine the mechanism by which C2 of BCTV induce genes, we performed transgenic Arabidopsis expressing BCTV C2. We are interesting to study the difference between the wild type and two mutants. -10 days old plants were grown in MS at 24 degrees in long day. Project description:affy_aba_ath1 - affy_aba_ath1 - In order to identify new genes implicated in ABA signalisation pathways, we have carried out a screen for suppressor mutants of the ABA deficient mutant aba3-1. Infrared thermography was used to identify plants whose cold leaf phenotype was reverted. The transcriptome analysis of four recessive suppressor mutants was undertaken to determine which genes are differentially expressed compared to the wild type or aba3-1. Comparison of control lines and transgenic lines expressing C2 in basal conditions and after treatment with 50uM MeJA for 10 hours. C2 lines are expressing geminivirus C2 under a 35S promoter. The jumoni proteins can remove the methyl group from lysine or arginine residues of histones. The aim of this project is to compare gene expression levels between mutants of two jumonji genes and wild type at seedling stage (12 d old).-- Seeds were sterilized in bleach, washed with ethanol, dried, dispersed on Murashige and Skoog (MS) 0.5X containing 2% sucrose and kept 5 days at 4Â°C. - After 12 days of growth under a cool white fluorescent light (120 Î¼mol m-2 s-1) and long day conditions (16h of light) in a growth chamber, the plants are harvested and stored at -80Â°C. -The RNA are extracted with the RNA Plant NucleoSpin (Machery-Nagel), quantified with Nanodrop and visualised on agarose gel. Geminivirus-encoded TrAP suppressor inhibits the histone methyltransferase SUVH4/KYP to counter host defense.

Project description:Transcriptional gene silencing (TGS) can serve as an innate immunity against invading DNA viruses throughout Eukaryotes. Geminivirus code for TrAP protein to suppress the TGS pathway. Here we identified an Arabidopsis H3K9me2 histone methyltransferase, Su(var)3-9 homolog 4 (SUVH4/KYP), as a bona fide cellular target of TrAP. TrAP interacts with the catalytic domain of KYP and inhibits its activity in vitro. TrAP elicits developmental anomalies phenocopying several TGS mutants, reduces the repressive H3K9me2 mark and CHH DNA methylation, and reactivates numerous endogenous KYP-repressed loci in vivo. Moreover, KYP binds to the viral chromatin, and controls its methylation to combat virus infection. Notably, kyp mutants support systemic infection of TrAP-deficient Geminivirus. We conclude that TrAP attenuates the TGS of the viral chromatin by inhibiting KYP activity to evade host surveillance. These findings provide new insight on the molecular arms race between host antiviral defense and virus counter defense at an epigenetic level.

Project description:The focus of this study was to identify changes in host gene expression induced by the transcription-dependent function of the viral AC2 protein, and induced by the interaction of AC2/C2 with SnRK1.2 (AtAKIN11). We used microarrays to identify changes in host gene expression induced by the transcription-dependent function of the geminivirus-encoded AC2 protein, and induced by the interaction of AC2 with a host serine-threonine kinase, SnRK1. Distinct classes of genes were up- and down regulated during this process. For experiment 1, Arabidopsis rosette leaves were infused with Agrobacterium capable of expressing full-length Cabbage leaf curl virus (CaLCuV) AC2 protein, a truncated version of AC2 lacking the C-terminal 29 amino acids (AC2del), or an empty vector control (530). In experiment 2, Arabidopsis rosette leaves were infused with Agrobacterium capable of expressing full-length Spinach curly top virus C2 protein, a truncated version of C2 lacking the C-terminal 29 amino acids (C2del), antisense (as) SnRK1.2 (AsAKIN11), or an empty vector control (530). Infused Arabidopsis rosette leaves were selected at 1, 2 or 3 days post-infusion for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain homogeneous populations of plants at each time point in order to increase the temporal resolution of expression profiles. To that end, we isolated total RNA from four individual plants, for three independent sets of plants infused with the different constructs. This resulted in three independent samples per treatment per time point.

Expression pattern in HaHB1 overexpressor plants under normal growth conditions

Project description:Plants deal with cold temperatures via different signal transduction pathways. To elucidate the molecular mechanisms by which HaHB1 conferred tolerance to cold temperatures, we performed an exploratory transcriptome analysis comparing RNAs of 3-week-old transgenic and WT plants. Several transcripts encoding glucanases and chitinases were induced. Notably, there was not a considerable overlap with CBF regulon genes previously described in cold tolerance molecular mechanism. We propose that HaHB1 is involved in plant cold tolerance via the induction of proteins able to stabilize cell membranes and inhibit ice growth. Transgenic vs WT experiment under normal conditions. Two independent transgenic lines. Two biological replicates for each line.
