Project description:affy_ralstonia_medicago - Ralstonia solanacearum is the causal agent of the devastating bacterial wilt disease. Its infection process was studied with an in vitro inoculation procedure on intact roots of Medicago truncatula. The pathosystem involved susceptible A17 and resistant F83005.5 M truncatula lines infected with the pathogenic strain GMI1000. The mutant A17 line, Sickle, which showed a resistant phenotype was also part of the experiment. To identify host signaling pathway triggered by R. solanacearum infection with a focus on the involvment of ethylene, we used the Medicago Affymetrix array to monitore the expression profiles and the molecular process associated with initial symptoms development (12hpi) and colonization (72hpi). In order to maximize chances to observe differential gene expression, RNA samples were extracted from the root infection zone (root tips) -Three Medicago truncatula lines, A17, F83005.5 and sickle were inoculated with GMI1000 Ralstonai solanacearum strain (107 cfu/ml). RNA were extracted from root extremities (1 cm above the root tip) at time 0, 12h and 72h post inoculation. Project description:affy_hypoctemp_medicago - The characterization of several genotypes of the model Legume Medicago truncatula showed a genetic variability for germination and hypocotyl heterotrophic growth at low temperature and optimal temperature. The most important contrast was between the accessions Jemalong A17 and F83005.5. In order to find genes differently expressed between temperatures and genotypes, the present work focuses on transcriptome profiling during hypocotyl heterotrophic growth under low (10°C) and optimal (20°C) temperature conditions for both genotypes. We used Jemalong A17 and F83005.5 seeds coming from the Medicago truncatula Biological Resource Center in Montpellier, produced in 2006. Experiments were performed in the dark to mimic pre-emergence growth. Pots (6.5 cm diameter, 10 cm high) were incubated in growth chambers either at 10°C or 20°C. They were filled with 500g sand and 100ml of a nutrient solution for young seedlings growth (Saglio and Pradet, 1980). Per pot, five scarified seeds were sown at 1.5 cm depth. Seedlings were harvested at three times: 35°Cd (degree day), 50°Cd and 100°Cd. At each timepoint, about 50 seedlings were harvested and hypocotyl lengths was measured. Project description:affy_agro-bi_medicago - Identification of genes from the model legume Medicago truncatula whose expression is affected by the plant nitrogen status, with or without inoculation with the symbiotic bacteria Sinorhizobium meliloti. - Comparison of the supernodulant, nitrogen-insensitive, sunn-2 mutant with the A17 wild type genotype.-The plant root systems of plants were split into two parts, each one being installed in a separate compartment. For the â??Sâ?? treatment, one part was supplied with 10 mM NH4NO3 while the other part was supplied with a nitrogen-free nutrient solution. For the â??Lâ?? treatment, one part was supplied 0.5mM NO3- and the other part was supplied with the nitrogen free solution. Eight biological materials (designated AL, AS, SL, SS, IL, IS, NIL, NIS), with three biological repeats for each, were collected and analyzed. Project description:The involvement of ROS in the legume – Rhizobium symbiotic interaction has been highlighted (Santos et al., 2001; Rubio et al., 2004). This interaction is characterized by the formation of a new organ on the root, the nodule and by the penetration, in parallel, of the bacteria into the root tissue via an infection thread (IT) (Parniske and Downie, 2003; Gage, 2004). H2O2 production has been shown in ITs during the Medicago – Sinorhizobium meliloti interaction (Santos et al., 2001). Moreover, S. meliloti mutants impaired in H2O2 detoxification mechanism possess in planta symbiotic phenotypes. As H2O2 production is known to orchestrate plant gene expression, our goal is focused on identifying the H2O2 regulated genes in the symbiotic process. We focus our analysis in the M. truncatula transcriptome as we are characterising the S. meliloti H2O2 transcriptome. -M. truncatula seedlings were grown in axenic condition on modified Fahraeus medium during seven days. Then they were transferred on new plates supplemented either with dimethyl sulfoxide (DMSO, mock treatement) or diphenylene iodonium (DPI, 10 µM). Twenty four hours later, they were inoculated either with water (mock inoculation; DMSO-H2O and DPI-H2O) or wild type S. meliloti 2011 strain (DMSO-INOC and DPI-INOC). Roots without apexes were harvested 48 hours after inoculation. Project description:affy_aba_ath1 - affy_aba_ath1 - In order to identify new genes implicated in ABA signalisation pathways, we have carried out a screen for suppressor mutants of the ABA deficient mutant aba3-1. Infrared thermography was used to identify plants whose cold leaf phenotype was reverted. The transcriptome analysis of four recessive suppressor mutants was undertaken to determine which genes are differentially expressed compared to the wild type or aba3-1. Project description:Plant apoplast is the first hub of plant-pathogen communication. Indeed, during interaction, pathogen effectors are recognized by plant defensive proteins and cell receptors and several signal transduction pathways are activated. Within this first contact, a defence response is triggered by the plant involving several extra and intracellular proteins. In grapevine, little is known about the communication between cells and apoplast in the context of plant-pathogen interactions, also apoplast dynamics, particularly considering the role of apoplastic proteins in response to pathogens still remains a blackbox. In this study we focused on 6 hours after Plasmopara viticola inoculation to evaluated grapevine proteome modulation both in the apoplastic fluid (APF) and total leaf tissue. Plasmopara viticola secretome was also assessed enabling a deeper understanding of plant and pathogen communication. Our results showed that oomycete recognition, plant cell wall modifications, ROS signalling and disruption of oomycete structures are triggered in Regent after P. viticola inoculation. Also, our results highlight a strict relation between the apoplastic pathways modulated and the proteins identified in ‘Regent’ total leaf proteome. On the other hand, P. viticola proteins related to growth/morphogenesis and virulence mechanisms were the most predominant. This pioneer study highlights the early dynamics of extra and intracellular communication between grapevine and defence activation leading to the successful establishment of an incompatible interaction
