Transcription profiling of thymus from ovariectomized mice to compare the effects of estrogen and its analog genistein on thymus.

ABSTRACT: Compared the effects of Estrogen and its analog Genistein on mouse thymus. Endogenous estrogen were minimized by removal of ovaries. Samples were collected at 4 time points: 0 days, 2 days, 3 days and 6 days from the start of the treatment. Two biological replicates at each time point. Experiment Overall Design: Control samples were from the mice that were untreated and at 0 day since treatment. Two treatments (Estrogen injection and Genistein diet) and three time points. Two replicated samples at each time point and each treatment. Total 14 samples.

Estrogen regulates thymic development and involution and modulates immune function. Despite its critical role in thymus, as well as in autoimmune disorders, the mechanism by which estrogen affects the thymus is not well understood. We previously reported that the estrogenic soy isoflavone genistein, as well as 17beta-estradiol (E2), could induce thymic involution, but genistein effects were only partially mediated through estrogen receptors. To provide insights into mechanisms of estrogenic effe  ...[more]
